摘  要:目的探讨不同治疗方案对消化道肿瘤患者外周血单个核细胞(PBMCs)中髓源性抑制细胞(MDSCs)亚群水平的影响。方法选取郑州大学第二附属医院消化道肿瘤患者40例和健康体检者10例,收集其化疗、手术、手术联合化疗前、后的外周血标本,用流式细胞术检测消化道肿瘤患者外周血粒细胞样MDSCs(G-MDSCs)和单核细胞样MDSCs(M-MDSCs)的表达,ELISA检测其功能因子Arg-1和iNOS。结果消化道肿瘤患者外周血中G-MDSCs和M-MDSCs的水平显著高于健康对照组。化疗、手术及联合治疗后外周血中G-MDSCs和M-MDSCs水平均降低,化疗、手术后MDSCs释放iNOS下降。G-MDSCs在有淋巴结转移、肿瘤直径≥5 cm的患者中较高;M-MDSCs在分化程度低的患者中较高。结论消化道肿瘤患者外周血中G-MDSCs和M-MDSCs亚群水平升高,手术、化疗以及二者联合可降低MDSCs亚群水平,并且降低MDSCs功能。G-MDSCs与肿瘤直径、淋巴结转移有关,M-MDSCs与分化程度有关。This study was performed to investigate the effect of different treatment regimens on the level of myeloid-derived suppressor cells (MDSCs) subsets in peripheral blood mononuclear cells (PBMCs) of patients with digestive tract cancers. Forty patients and l0 healthy people were recruited from the Second Affiliated Hospital of Zhengzhou University. Peripheral blood samples were collected before and after chemotherapy, surgery or surgery combined with chemotherapy. Granulocytic MDSCs (G-MDSCs) and monocytic MDSCs (M-MDSCs) levels were detected by flow cytometry; while Arg-1 and iNOS released by MDSCs were detected by ELISA. Data showed that the levels of G-MDSCs and M-MDSCs in peripheral blood of patients were significantly higher than those in healthy people, and decreased after treatment; the level of iNOS released by MDSCs was decreased after surgery or chemotherapy. Moreover, G-MDSCs level was positively related to lymph node metastasis and tumor size (35 cm), while M-MDSCs level was negatively related to cancer differentiation. In conclusion, MDSCs subsets in peripheral blood of digestive tract tumor patients are increased and can be reduced by chemotherapy and/or surgery. Levels of iNOS of MDSCs are decreased after surgery or chemotherapy. G-MDSCs are associated with tumor size and lymph node metastasis, while M-MDSCs are related to the degree of tumor differentiation.
