益智仁和砂仁有效成分对岗田酸诱导阿尔兹海默病细胞模型的作用研究

摘　　要：目的:探讨益智仁和砂仁有效成分对岗田酸诱导阿尔兹海默病模型细胞的作用。方法:采用CCK8法确定1~48 h内冈田软海绵酸(okadaic acid,OA)对高分化PC12细胞的作用时间,原儿茶酸和乙酸龙脑酯的作用浓度,随之将其分为空白对照组,模型对照组,原儿茶酸组和乙酸龙脑酯组,检测细胞中p-tau含量和观察细胞形态变化。结果:175 nmol·L^(-1)浓度OA的适应造模时间是48 h,原儿茶酸和乙酸龙脑酯的LD50分别是0.31 nmol·L^(-1)和0.80 nmol·L^(-1)。与空白对照组比较,模型对照组的p-tau显著增加(P<0.01),说明OA诱导p-tau细胞模型造模成功;与模型对照组比较,原儿茶酸组和乙酸龙脑酯组的p-tau显著减少(P<0.01),细胞形态的损伤减少。结论:原儿茶酸和乙酸龙脑酯对OA诱导阿尔兹海默病模型细胞有一定的作用。Objective： To study the effect of the component of Fructus Alpiniae Oxyphyllae and Amomum Villosum on Alzheimer＇s disease（ AD） cell model induced by Okadaic Acid（ OA）. Method： The effect of OA（ 175 nmol·L^-1） within 48 hours was detected by CCK8 method. And the concentration of Protocatechuic Acid and Bornyl Acetate for high differentiated PC12 cell in 48 hours was detected by CCK8 method. Then it can be divided into blank control group,model control group,Protocatechuic Acid treated-group and Bornyl Acetate treated-group. P-tau content and morphological change was tested. Results： The appropriate time of OA was 48 hours. The appropriate concentration of Protocatechuic Acid and Bornyl Acetate in 48 h was 0. 31 nmol·L^-1 and 0.80 nmol·L^-1. P-tau in the model control group increased significantly compared with that in the blank control group（ P〈0. 01）.It means that p-tau cell model induced by OA was successfully built. P-tau levels of Protocatechuic Acid treated-group and Bornyl Acetate treated-group significantly decreased（ P〈0. 01） and cell morphology damage reduced compared with those in the model control group. Conclusion： Protocatechuic Acid and Bornyl Acetate have an effect on Alzheimer＇ s disease cell model induced by OA.
