摘  要:目的观察川芎嗪(TMP)对兔良性胆管狭窄(BBS)胆管成纤维细胞(MF)增殖及细胞外基质代谢的影响。方法对家兔采用切开再吻合方法建立BBS模型并获取MF,经细胞鉴定后将MF随机分为两组,观察组给予0.04、0.2、1.0 mg/m L的TMP,对照组不处理。干预48 h后,采用CCK-8法观察细胞增殖情况(OD值),分别采用Real-time PCR法检测细胞中的Ⅰ型胶原(col-Ⅰ)、基质金属蛋白酶9(MMP-9)及基质金属蛋白酶抑制因子1(TIMP-1)mRNA。结果观察组予0.04、0.2、1.0 mg/m L的TMP处理48 h时,其OD值均低于对照组(P均<0.05);观察组予0.2、1.0 mg/mL的TMP处理48 h时,其col-Ⅰ、TIMP-1 mRNA相对表达量均低于对照组(P均<0.05),MMP-9 mRNA相对表达量高于对照组(P均<0.05)。结论 TMP可通过影响细胞外基质代谢抑制MF增殖,从而对BBS起到治疗作用。Objective To investigate effects of tetramethylpyrazine(TMP) on cell proliferation and extracellular matrix metabolism in the benign biliary stricture(BBS) model fibroblasts(MF) of rabbits.Methods MF were acquired from rabbit BBS model and identified,then were divided into two groups: the observation group(treated with 0.04,0.2,and1.0 mg/m L TMP) and control group.After 48-hour intervention,the cell proliferation(OD value) was investigated by CCK-8; the mRNA expression levels of matrix metalloproteinase 9(MMP-9),tissue inhibitor of matrix metalloproteinases-1(TIMP-1),and collagen-I(col-I) were investigated by real-time PCR.Results The OD value in the observation group treated with 0.04,0.2,and 1.0 mg/m L TMP for 48 h was lower than that in the control group(all P 〈 0.05).The relative mRNA expression of col-Ⅰand TIMP-1 in the observation group treated with 0.2 and 1.0 mg/m L TMP for 48 h was lower than that of the control group(all P 〈 0.05),and the relative expression of MMP-9 mRNA was higher than that of the control group(P 〈 0.05).Conclusion TMP can play a therapeutic role in BBS by affecting the extracellular matrix metabolism and inhibiting MF proliferation.
