摘  要:目的在前期研究中进行全基因组表达谱检测发现在雄激素受体(androgenreceptor,Ar)基因睾丸支持细胞特异性敲除小鼠(s—Ar/y)睾丸组织中干扰素刺激因子15(interferon—stimulatedgene15,Isg15)基因表达增高。研究的目的是了解睾丸组织中雄激素及其受体对Isg15基因表达的影响作用。方法采用RT—qPCR和Western印迹等方法比较野生型小鼠和s—Ar由小鼠睾丸组织中Isg15基因表达量。通过免疫组织化学和荧光染色两种方法检测Isg15蛋白在睾丸组织中的分布。结果在s—Ar由小鼠睾丸组织中tsg15基因表达量比在野生型小鼠中显著升高,Isg15蛋白主要表达于精母细胞。结论睾丸支持细胞特异性敲除加基因使精母细胞中Isg15基因表达异常增高。Objective The expression of interferon-stimulated gene 15 (Isg15) was found to increase in the testes of Sertoli cell specific Ar knockout (S-Ar-/y) mice. This study aimed to exam- ine the effect of androgen and its receptor on the expression of Isg15. Methods RT-qPCR and Western blotting were used to detect the expression of Isgl5 in wide-type mice and S-Ar/y mice and the difference was compared. The distribution of Isg15 protein in mouse testis was measured by im- munohistochemistry and immunofluorescence. Results The expression of Isg15 gene was signifi- cantly higher in S-At-/y mice than that in wild-type mice, and the Isg15 protein was mainly expressed in spermatocytes. Conclusion Sertoli cell specific knockout of Ar gene Results in abnormally high expression of Isg15 gene in spermatocytes.
