摘 要:目的:探讨孕鼠手机辐射暴露对子鼠前额叶皮质中5-HT_(2C)受体表达水平的影响。方法:27只SPF级SD孕鼠平均随机分为3组:A组(不辐射)、B组(辐射6 h/d,21 d)和C组(辐射24 h/d,21 d),每组9只孕鼠;待子鼠出生后每组随机留养72只,在出生后第1 d、7 d、30 d和60 d取子鼠前额叶皮质。运用免疫组织化学方法标识5-HT_(2C)受体表达,并通过图像分析软件对阳性表达进行IOD值分析;运用实时荧光定量PCR法检测5-HT_(2C)受体mRNA的相对表达水平。结果:5-HT_(2C)受体阳性标识主要集中在神经元胞质内,阳性表达的IOD值和实时荧光定量PCR法检测5-HT_(2C)受体表达水平的结果是一致的:与A组相比,B、C两组的表达水平均显著升高(P<0.05);且C组水平高于B组(P<0.05)。结论:孕鼠手机辐射暴露可导致子鼠前额叶皮质5-HT_(2C)受体表达水平升高。Objective: In this study, the aim was to investigate the effect of pregnant rats exposure to mobile phone radiation on offspring rats'5-HT2c receptor expression in prefrontal cortex. Methods: Twenty-seven specific pathogen free Sprague-Dawley pregnant rats were randomly divided into three equal groups:group A (normal breeding), group B ( radiationg for six hours per day during all the pregnancy) and group C ( radiationg for twenty-four hours per day during all the pregnancy), then each group consisted of nine pregnant rats. Keeping seventy-two offspring rats randomly in each group, and taking the cerebral pretrontal cortex respectively at the first day after birth, seven days after birth, thirty days after birth, sixty days after birth. Immunohistochemistry method was used to measure the locationg and expression level of 5-HT2c receptor in prefrontal cortex, and the quantitative value of IOD on positive expression was analyzed by the image analysis software. Real-time fluorescence quantitative PCR method was used to speculate the relative expression level of 5-HT2c receptor (mRNA). Results: The positive identification on 5-HT2c receptor was mainly expressed in the cytoplasm of neuron. The results of the immunohistochemical method and the real-time fluorescence quantitative PCR method were consistent. Compared with the A group, the expression levels on 5-HT2c receptor of the B group and the C group were significantly higher ( P 〈 0. 05 ), and the C group was higher than that of B group ( P 〈 0.05 ). Conclusion: Pregnant rats exposure to mobile phone radiation could induce higher expreession of 5-HT2c receptor in prefrontal cortex of offspring rats.
