摘  要:对不同酸枣仁提取与纯化工艺进行对比,以提高酸枣仁皂苷含量和纯度。以酸枣仁皂苷A的含量为各浸膏皂苷含量及纯度的评价指标,采用显色法/紫外-可见分光度法进行检测。正丁醇萃取后,采用氨洗或硅胶分配色谱法进行纯化,计算各提取纯化方法所得浸膏的皂苷含量和纯度。回流提取法中,其提取、正丁醇萃取物、氨洗或硅胶分配色谱出膏率分别为16.28%,30.52%,54.01%,54.06%,纯度分别为17.91%,35.43%,54.12%,63.92%;微波提取法对应出膏率分别为12.24%,26.09%,57.60%,55.97%,纯度分别为15.49%,38.11%,58.01%,65.10%。回流法出膏率较微波提取法高,但微波提取物最终纯度较高;硅胶分配色谱法纯化效果优于氨洗;得到纯度65.10%的酸枣仁皂苷。Different extraction and purification processes were used to improve Jujuboside content and purity. In evaluation of Jujuboside extraction and purity,chromogenic assay and UV-visible spectrophotometry were used. The extraction of Jujuboside obtained by reflux and microwave methods was concentrated by n-butanol extraction,ammonia washing,and silica gel chromatography. In extraction of reflux,the saponin content of the extraction,n-butanol extraction,ammonia washed cream and silica partition chromatography were 16. 28%,30. 52%,54. 01%,and 54. 06%,respectively. And,the purities were 17. 91%,35. 43%,54. 12%,and 63. 92%,respectively. In comparison,the saponin content of microwave-assistant extraction method were 12. 24%,26. 09%,57. 60% and 55. 97%,respectively. Their purities were 15. 49%,38. 11%,58. 01%,and 65. 10%,respectively. The saponin content of reflux extraction is higher than microwave extraction. However,the obtained purity of reflux extraction is lower than microwave. Purification of silica partition chromatography is better than ammonia washing. The Jujuboside purity of 65. 10%was obtained in the study.
