摘　　要：目的:检测染料木黄酮对小鼠颈总动脉组织磷酸化蛋白激酶B(p-Akt)表达水平的影响,并探究染料木黄酮对于小鼠颈总动脉粥样硬化的预防及治疗作用。方法:实验选用50只6周龄的SPF级纯种雄性C57BL6小鼠,随机分为5组,分别为空白对照组、模型组、阿托伐他汀钙组、低浓度染料木黄酮组和高浓度染料木黄酮组。颈总动脉套管后喂养14周处死小鼠,血清检测血脂4项,H-E染色观察形态学变化,油红O染色观察脂滴含量,蛋白质印迹和免疫荧光检测p-Akt蛋白含量。结果:空白对照组,高、低浓度染料木黄酮组和阿托伐他汀钙组的血脂水平明显低于模型组。组织病理学观察表明,除模型组外,其余各组颈动脉结构均无斑块形成,且脂滴含量较少。蛋白质印迹和免疫荧光检测分析显示,模型组p-Akt表达水平显著高于染料木黄酮组、阿托伐他汀钙组和空白对照组。结论:染料木黄酮可通过下调p-Akt进而减轻小鼠颈总动脉粥样硬化的形成。Objective： To investigate the expression of phosphorylated protein kinase B （p-Akt） in common carotid artery tissue of mice, and to investigate the preventive and therapeutic effects of genistein on carotid atherosclerosis in mice. Methods： Fifty male C57BL6 mice with 6-week old of SPF class were randomly divided into control group, model group, atorvastatin group, low genistein group, and high genistein group. Mice were fed for 14 weeks after carotid artery cannula. Blood serum was used to test low density lipoprotein （LDL）, triglyceride （TG）, total cholesterol （TC） and high density lipoprotein （HDL）. Meanwhile, H-E staining, oil red staining, Western blotting and immunofluorescence were used to detect p-Akt of common carotid artery in mice. Results： Compared with the control group, blood lipid of high-genistein group, low- genistein group and atorvastatin group was lower than model group. Histopathological observation showed that plaque formation was only found in model group, and there were more lipid droplets in the blood vessel of model group than the other groups. Western blotting showed that the expression of p-Akt in model group was significantly higher than genistein group, atorvastatin group, and control. The trend of p-Akt by immunofluorescence was similar to Western blotting. Conclusion： Genistein can reduce the formation of carotid atherosclerosis by downregulating p-Akt in mice.
