摘 要:目的:探讨异甘草素对人肝癌HepG2细胞的增殖及超微结构的影响。方法:采用CCK8法检测不同浓度异甘草素对HepG2细胞的增殖作用。透射电镜观测HepG2细胞形态变化。流式细胞仪检测细胞凋亡率。结果:CCK8法检测到异甘草素能够显著抑制HepG2细胞的增殖,并呈浓度和时间依赖性。透射电镜结果显示,异甘草素可引起细胞超微结构的明显改变。HepG2细胞经异甘草素作用24h和48h后,细胞凋亡率显著增加。结论:异甘草素对人肝癌HepG2细胞的增殖具有明显的抑制作用。Objective: To investigate the effect of isoliquiritigenin on proliferation and ultrastructure of human hepatocarcinoma HepG2 ceils. Methods: The growth inhibition was detected by cell counting kit 8 (CCK8). The ultrastructure of HepG2 cells was observed by transmission electron microscope. Apoptosis rate was determined by flow cytometry. Results : CCK8 method detected that isoliquiritigenin could inhibit the proliferation of HepG2 cells in a concentration and time dependent way. Typical morphological alterations were observed by transmission electron microscope on the isoliquiritigenin treated cells. Apparent enhancement in apoptosis rate was detected using flow cytometry after HepG2 ceils had been exposed to the isoliquiritigenin for 24 hours and 48 hours, respectively. Conclusion: Isoliquiritigenin has a significant inhibitory effect on proliferation of human hepatocarcinoma HepG2 cells.
