摘  要:本试验的目的是采用高通量测序技术,研究北京动物园3只亚成体大熊猫肠道细菌的多样性,比较分析在其刚从成都回北京动物园饲养(14月龄)和饲养2个月后(16月龄)的粪便菌群差异,以及3只个体之间的差异。共采集新鲜粪便样品19份,提取DNA,然后用Illumina MiS eq平台测序分析。结果表明:3只亚成体大熊猫粪便细菌,在门水平上主要是变形菌门(Proteobacteria),平均占55.5%,厚壁菌门(Firmicutes)平均占42.5%;在属水平上,大肠志贺氏杆菌属(Escherichia shigella)平均占50.1%,狭义梭菌属(Clostridium sensu stricto)平均占13.8%,链球菌属(Streptococcus)平均占11.7%。在门到属的各级分类水平上,14月龄和16月龄的粪便菌群除了乳球菌属(Lactococcus)外,差异不显著(P>0.05);3只个体之间菌群差异不显著(P>0.05)。We studied intestinal bacterial diversity of three sub-adult giant panda of the Beijing Zoo by High-Throughput Sequencing Technology. We analyzed the differences between diversity on the arrival of the pandas at Beijing Zoo( 14 months age)and after two months of feeding at Beijing Zoo(16 months age). We also compared diversity between the three pandas. Nineteen fresh fe- cal samples were collected. DNA was extracted and analyzed using the Illumina MiSeq platform. The gut bacteria were mainly Proteobacteda, which accounted for an average of 55. 5% and the Firmicutes accounted for an average of 42.5% at the phylum level. Escherichia shigella accounted for an average of 50, 1%, Clostridium sensu stficto for an average of 13. 8%, and Streptococcus for an average of 11.7% at the genus level. At taxonomic levels from phylum to genus, the fecal microfiora at 14 and 16 months were similar( P〉 0. 05). Except for Lactococcus, bacterial popu- lations were similar among three pandas ( P 〉 0. 05).
