摘 要:目的:探讨NAC-1在自噬介导的卵巢癌细胞顺铂耐药中的作用。方法:Western blot及流式细胞术检测顺铂诱导的耐药卵巢癌细胞(SKOV3/DDP)自噬及凋亡过程中,NAC-1蛋白的表达情况。采用siRNA下调NAC-1表达,检测其对SKOV3/DDP细胞自噬及凋亡的影响,并检测其对顺铂耐药性的影响。结果:顺铂处理SKOV3/DDP细胞后,细胞自噬及凋亡水平升高,NAC-1蛋白表达升高。siRNA下调NAC-1基因后,顺铂处理SKOV3/DDP细胞的自噬水平降低,凋亡升高,细胞顺铂耐药性降低。结论:NAC-1基因可能通过调节自噬参与卵巢癌细胞顺铂耐药,降低NAC-1基因表达有助于提高顺铂对卵巢癌细胞的杀伤作用。Objective: To investigate the role of NAC-1 in autophagy mediated cisplatin resistance in ovarian cancer cells. Methods: Autophagy and apoptosis of cisplatin-induced chemoresistant ovarian cancer cells( SKOV3/DDP) were detected by Western blot and Flow cytometry.The expression of NAC-1 protein was detected.The effect of NAC-1 inhibition by siRNA on autophagy,apoptosis and resistance of SKOV3/DDP cells were detected. Results: Cisplatin could lead to increased autophagy and apoptosis of SKOV3/DDP,while NAC-1 protein expression was increased.Downregulation of NAC-1 by siRNA could decrease autophagy and increase apoptosis in SKOV3/DDP cells treated with cisplatin. Meanwhile,cisplatin resistance was decreased.Conclusion: NAC-1 gene may participate in cisplatin resistance of ovarian cancer cells by regulating autophagy.Reducing the expression of NAC-1 gene may be used to improve the cytotoxicity of cisplatin on ovarian cancer cells.
