教学实验常用细菌鞭毛染色条件的研究

摘　　要：目的比较培养条件及制片方法对变形杆菌鞭毛染色效果的影响,找出最适条件。方法将变形杆菌接种于不同培养基,培养不同时间,采取不同孵化制片方法,控制单一变量,染色镜检,比较最佳染色条件。结果血琼脂培养基中培养的细菌鞭毛粗大清晰,染色效果最好。普通培养基培养16 h的细菌鞭毛长度合适,结构清晰,染色效果好。37℃温箱孵育10~15 min的细菌鞭毛展开充分,染色效果最好。结论血琼脂培养基是观察鞭毛染色的最适宜培养基;16 h是细菌在普通琼脂培养基中的最适宜培养时间;温箱孵育10~15 min是使鞭毛充分展开的最适宜孵化时间。Objective To compare the efects of diferent culture medium,different culture time and different production methods on the stainingof the bacterial flagellum. Methods Inoculate the Proteus in differentculture medium, cultured for different time, take different methods ofincubation, control single variable, comparing the optimum stainingconditions. Results The bacterial fagellum in the blood agar mediumis large and clear, the background is clean, the staining effect is thebest; the 16 hours’ bacterium is of medium size with clear andcomplete flagellum, whose microscopic examination look the best.Incubated in 37℃ for 10 to 15 minutes, the bacterial fagellum expandsfully, showing the best effect of staining. Conclusion The blood agarmedium is the best culture medium for the teaching and identifcation.The 16 hours is the best culture time of the bacteria in the common agarmedium; incubation 10 to 15 minutes is the best incubation time for thefull expansion of the bacterial fagellum.
