摘 要:目的:应用基因芯片技术对理冲生髓饮作用后人卵巢癌基因表达谱进行分析并且筛选差异表达基因,从信号转导方面和血管生成方面探讨中药复方理冲生髓饮对人卵巢癌作用机制。方法:建立Balb/C裸鼠卵巢癌皮下移植瘤模型,随机分为模型组和中药中剂量组。停药后将肿瘤组织分离。并且采用基因芯片技术,研究其表达图谱,从中筛选出中药中剂量组与模型组差异基因表达,探讨中药可能作用机制。结果:在分析差异基因的表达时,探索中药复方理冲生髓饮作用后,可能改变的信号通路有Jak-STAT信号通路、TNF信号通路、Nf-κb信号、HIF-1信号通路、趋化因子信号通路以及t细胞受体信号通路和胰岛素信号通路,BAG信号通路等。结论:基因芯片分析与卵巢癌相关差异基因12条;差异表达基因可能与肿瘤血管生成相关。Objective: To explore the anti ovarian cancer mechanism of compound Lichong Shengsui Yin(LSY) from the aspect of signal transduction and angiogenesis,based on the analysis of gene spectrum and the screening of discrepant gene expression with gene chip technology. Methods: Balb/C nude mice with implanted ovarian cancer were randomly divided into the model group and LSY group of medium-dose,tumor tissue was separated after the medication. Gene expression spectrum was studied to screened out the discrepant gene expression in LSY group with 4* 180 k Affymetrix OElnc RNA chip technique(m RNA32,776),thus to explore its possible anti-tumor mechanism. Results: Through analyzing the discrepant gene expression,possible signal pathways changed might involved signal pathways of Jak-STAT,HIF-1,TNF,Nf-κb,T cell receptors,chemotactic factor,insulin,and BAG. Conclusion: There are discrepant gene expression related to ovarian cancer,which may be associated with angiogenesis of the tumor.
