摘  要:目的:研究覆盆子对酒精戒断焦虑症的治疗作用,并探讨起对脑内海马组织中去甲肾上腺素的机制。方法:选取健康SD大鼠36只,体重220~250 g,随机分为6组,空白组、模型组、高剂量药物组、中剂量药物组、低剂量药物组、阳性药物组。除空白组外,其余5组每天腹腔内注射20%(w/v)酒精1.5 ml/100 g,共28天,制备酒精成瘾模型,然后灌胃不同剂量药物,每天一次,共给3天,最后一次给药1 h后,用高架十字迷宫测其焦虑行为,断头取脑,分离海马,用HPLC测海马中去甲肾上腺素的含量。结果:在高架十字迷宫中,与空白对照组相比,酒精戒断大鼠在开放臂的逗留次数、时间明显减少(P<0.01);与模型组相比,高剂量覆盆子提取物显著地增加其次数、时间(P<0.05),而中、低剂量覆盆子提取物作用效果不明显(P>0.05);HPLC检测显示与空白组相比,酒精成瘾大鼠海马中去甲肾上腺素的含量显著升高(P<0.01),与模型组相比,高剂量覆盆子可降低海马中去甲肾上腺素含量(P<0.05),中、低剂量覆盆子降低去甲肾上腺素含量的趋势不明显(P>0.05)。结论:覆盆子提取物能减轻酒精戒断时大鼠所表现出的焦虑样行为,其抗焦虑症的作用机制可能与降低大鼠脑内海马组织中去甲肾上腺素含量有关。Objective: To investigate the effect of raspberry on anxiety-like behavior and the levels of norepinephrine(NE) in hippocampus after ethanol withdrawal. Methods: 36 SD rats were randomly divided into the blank group,the model group,high-dose raspberry group,medium-dose raspberry group,low-dose raspberry group,and positive control group,with 6 rats in each group. Apart from the blank group,the other groups were given 20% ethanol through intraperitoneal injection for 28 days to establish the models. After modeling,the medication intervention was applied for three days. One hour after the final medication,the anxiety-like behavior was evaluated by using an elevated maze,the concentrations of NE in hippocampus were measured by HPLC method. Results: In the maze test,rats undergoing ethanol withdrawal stayed shorter in the open arms of the elevated maze(P < 0. 01); compared to the model group,the time was increased in the high-dose raspberry group(P < 0. 05); however,the medium-dose raspberry group and the low-dose raspberry group had no obvious effect(P > 0. 05). In terms of the content of NE,they were all increased except the blank group; after medication,they were decreased in the high-dose raspberry group compared to the model group(P < 0. 05),the medium-dose raspberry group and the low-dose raspberry group had no obvious effect(P >0. 05). Conclusion: The extract from raspberry can attenuate anxiety-like behavior induced by ethanol withdrawal through modulating NE in hippocampus.
