寒凝血瘀型子宫内膜异位症大鼠模型的制备

摘　　要：目的:病证结合的制备寒凝血瘀型子宫内膜异位症大鼠模型,并对该模型各项指标进行动态观察。方法:采用冰水浸泡法加皮下注射肾上腺素法先建立寒凝血瘀型大鼠模型,在该模型的基础上,采取自体子宫内膜移植配合冰水浸泡法建立大鼠模型,空白对照组只切开腹部,不移植内膜。每组选取6只大鼠,分别于手术后3天、5天、1周、2周、3周、4周、6周、8周及12周进行取材,观察大鼠中医证候表现及大体病灶外观,对异位组织行病理学观察,心尖采血检测凝血象及血流变。结果:1.冰水浸泡14天配合皮下注射肾上腺素法可以使大鼠具有中医"寒凝血瘀"的表现;2.除术后12周外,各组大鼠都具备寒凝血瘀证的表现,术后4周起大鼠出现了子宫内膜异位症的病理表现。结论:此种方法造模符合临床患者病机演变规律,同时该模型具有稳定性;该模型对寒凝血瘀型子宫内膜异位症的研究具有应用价值。Objective: To establish endometriosis model of blood stasis due to cold congealing in rats guided by the differentiation of disease and syndrome,and to carry out the dynamic observation of various indicators of the model. Methods:The model of blood stasis due to cold congealing was firstly established in rats with the method of ice water immersion and subcutaneous injection of adrenaline; on which basis,autologous endometrial transplants combined with ice water immersion was also applied,the blank control group was only intervened by abdominal incision without endometrium transplantation. 6 rats from each group was randomly selected 3 d,5 d,1 w,2 w,3 w,4 w,6 w,8 w,and 12 w respectively to observe the appearance and the syndrome manifestation of the rats,as well as to observe the pathology changes of heterotopic endometrium,blood was collected through apex cordis for coagulation test and hemorrheology test.Results: The method of 14-day ice water immersion with subcutaneous injection of adrenaline could make the rats have the syndrome manifestation of blood stasis due to cold congealing. Except the group of 12 w after the operation,rats in the other groups all showed the manifestation of blood stasis due to cold congealing; the rats in 4 w after the operation showed the pathological manifestation of endometriosis. Conclusion: This method is consistent with the evolution of pathogenesis of clinical patients,the model has stability and has application value in the study of endometriosis with blood stasis due to cold congealing.
