摘  要:【目的】为黔北麻羊种质资源的综合开发利用提供参考。【方法】应用DNA池结合PCR产物直接测序方法检测黔北麻羊DKK1基因CDS区及部分内含子的多态性,应用生物信息学方法对黔北麻羊DKK1基因的蛋白特性进行预测和分析。【结果】在黔北麻羊的DKK1基因CDS区及部分内含子序列中筛选出8个SNP位点,分别为第1内含子C1626T位点,第3内含子A3083G位点,编码区的T3000C、T3228C、T3238C、G3240A、T3271A和T3411C位点。其中,T3271A为错义突变,导致半胱氨酸(Cys)变为丝氨酸(Ser),其他编码区的SNP位点均为同义突变。DKK1基因的CDS区包含有1条长789 bp的核苷酸序列,编码262个氨基酸,蛋白分子量为28 349.17 D;DKK1基因编码的整个多肽链表现为亲水性,并具有信号肽序列,剪切位点最有可能位于第31~32位氨基酸,在第15~37位氨基酸间存在1个典型的跨膜结构域;其二级结构主要由α-螺旋、β-折叠和无规则卷曲组成,属于mixed型蛋白;黔北麻羊DKK1基因编码的氨基酸与普通山羊、绵羊和牛等物种均具有较高同源性,基因系统进化情况与其亲缘关系基本一致。【结论】DKK1基因在进化上较保守,其可作为动物骨骼生长发育研究和相关疾病鉴定的重要候选基因。【Objective】This paper aims to provide a reference for intensive research of Qianbei Ma Goat comprehensive exploitation and utilization of its germplasm resources.【Method】The polymorphism of CDS region and part introns of Qianbei Ma Goat DKK1 gene is detected by using DNA pool and direct sequencing of PCR products and the protein characteristics of Qianbei Ma Goat DKK1 gene is forecast and analyzed by the bioinformatic software.【Result】8 SNP sites are screened from sequences of CDS region and part introns of Qianbei Ma Goat DKK1 gene and there are C1626 T site in the first intron,A3083 G site in the third intron and six SNP sites of T3000 C,T3228 C,T3238 C,G3240 A,T3271 A and T3411 C in CDS region. T3271 A is missense mutation,leading to Cys → Ser. The mutation of other SNP sites in CDS region is synonymous mutation. The CDS region of DKK1 gene contains a 789 bp nucleotide sequence encoding 262 amino acids with 28349. 17 D protein molecular weight. The encoded polypeptide chain with signal peptide sequence is hydrophilicity. The cleavage site is at the 31^(th)-32^(th) amino acids most possibly and there is a typical transmembrane domain among the 15^(th)-37^(th) amino acids. The biomolecular structure of the polypeptide chain is composed of α-heli,β-sheet and random coil,which belongs to the mixed protein. The amino acid encoded by Qianbei Ma Goat DKK1 gene has a higher homology with common goat, sheep, cattle and so on. The systematic evolution of DKK1 gene is consistent with its genetic relationship basically.【Conclusion】The DKK1 gene can be used as an important candidate gene for research of animal bone growth and development and identification of relative diseases because the evolution of DKK1 gene is conservation.
