摘 要:目的研究淫羊藿苷(ICA)对大鼠精子质量和精核蛋白(TNBP)作用的影响。方法将80只SD雄性大鼠分为对照组(蒸馏水3 m L灌胃60 d)、模型组(先用腺嘌呤25 mg/100 g和蒸馏水3 m L配置后灌胃30 d后,再以蒸馏水3 m L灌胃30 d)、ICA低剂量组(先用腺嘌呤25 mg/100 g和蒸馏水3 m L配置后灌胃30 d后,再以ICA 1.8 g/kg和蒸馏水3 m L灌胃30 d)和ICA高剂量组(先用腺嘌呤25 mg/100 g和蒸馏水3 m L配置后灌胃30 d后,再以ICA 3.6 g/kg和蒸馏水3 m L配置后灌胃30 d),每组20只。观察大鼠睾精核蛋白表达和精子质量变化。结果模型组、ICA低剂量组、ICA高剂量组精子密度、活率和活力及大鼠精核蛋白(RP)值均显著低于对照组,总碱性蛋白(TH)、TH/RP值均显著高于对照组,差异均有统计学意义(P<0.05);ICA低剂量组大鼠精子密度和活力与模型组比较,差异均无统计学意义(P>0.05)。ICA高剂量组精子活率、活力及RP值均显著高于模型组,TH、TH/RP值均显著低于模型组,差异均有统计学意义(P<0.05)。结论高剂量ICA可以改善精子活力和TNBP含量,但不能显著增加精子密度。Objective To study the effect of icariin(ICA)on sperm quality and spermatid unclear protein of rats.Methods A total of 80 male SD rats were divided into the control group(gavage with 3 m L distilled water for 60 d),the modelgroup(gavage with 3 m L distilled water for 60 d after combination of 25 mg/100 g adenine and 3 m L distilled water for 30 d),theICA low-dose group(gavage with 1.8 g/kg ICA and 3 m L distilled water for 30 d after combination of 25 mg/100 g adenine and3 m L distilled water for 30 d)and ICA high-dose group(gavage with 3.6 g/kg ICA and 3 m L distilled water for 30 d after combina-tion of 25 mg/100 g adenine and 3 m L distilled water for 30 d),20 cases in each group. The expression of spermatid unclear proteinand changes of sperm quality of the rats in all the groups were observed. Results The rats′ sperm density,motility rate,vitalityrate and RP value in the model group,ICA low-dose group and ICA high-dose group were obviously lower than those in the con-trol group,but the TH value and TH/RP ratio were prominently higher than those in the control group,the difference had statisti-cal significance(P<0.05). There were no statistically significant difference on rats′ sperm density and vitality rate between theICA low-dose group and the model group(P>0.05). The sperm′ s motility rate,vitality rate and RP value in the ICA high-dosegroup were remarkably higher than those in the model group,but the TH value and the TH/RP ratio were prominently lower thanthose in the model group,the difference had statistical significance(P<0.05). Conclusion High-dose ICA can improve the vi-tality rate of sperm and content of TNBP,but it cannot significantly increased the sperm density.
