摘 要:探讨羽衣甘蓝乙醇提取物对过氧化氢(H_2O_2)诱发小肠上皮Caco-2细胞氧化应激损伤的保护作用。MTT法测定干预后细胞的生存率,比色法测定细胞内超氧化物歧化酶(Superoxide dismutase,SOD)、过氧化氢酶(Catalase,CAT)和谷胱甘肽过氧化酶(Glutathione peroxidase,GSH-Px)水平。硫代巴比妥酸法(Thiobarbituric acid reactive substance,TBARS)测定细胞内氧化损伤标志物丙二醛(Malondiadehycle,MDA)含量。活性氧(ROS)用2’,7’-二氢二氯荧光黄双乙酸钠(Dichloro-dihydro-fluorescein diacetate,DCFH-DA)探针测定。酶联法(Enzyme linked immunosorbent assay,ELISA)法测定细胞内白介素(Interlukin,IL)-8的分泌量。羽衣甘蓝乙醇提取物能明显提升受损细胞的生存率及细胞内源性抗氧化物酶(SOD、CAT和GSH-Px)的活性。降低受损细胞内ROS与MDA水平,并抑制IL-8分泌。研究结果提示羽衣甘蓝乙醇提取物能显著改善H_2O_2诱发Caco-2细胞所发生的氧化应激损伤。To investigate the effect of Brassica Oleracea ethanol extracts(BOEE) on oxidative stress in intestinal epithelial Caco-2 cells. Cell viability was determined by MTT assay. The cellular levels of superoxide dismutase(SOD), catalase(CAT) and glutathione peroxidase(GSH-Px) were determined by commercial assay kits according the manuscripts. The cellular levels of malondiadehycle(MDA) and reactive oxygen specisis(ROS) were determined by thiobarbituric acid reactive substance(TBARS) and dichloro-dihydro-fluorescein diacetate(DCFH-DA) assay, respectively. The levels of interlukin-8(IL-8) was determined used a enzyme linked immunosorbent assay(ELISA). BOEE was significantly increased the cell viability and activity of SOD, CAT and GSH-Px in cells. In addition, BOEE treatment was also effectively decreased the cellular levels of ROS and MDA, and also reduced the IL-8 secretion in H_2O_2 treated Caco-2 cells. These results suggested the BOEE exhibited a protective effect to against H_2O_2-induced oxidative stress in Caco-2 cells.
