摘  要:将高通量且无损的核磁共振氢谱(~1H-NMR)测试技术与多种多元统计分析方法相结合,探索从代谢组学角度进行阿胶原料来源鉴别。针对14个不同来源的固体动物胶样品,在相同条件下进行~1H-NMR图谱测试和图谱标准化处理,得到反映样品来源代谢组学特征的数据集,利用统计软件进行系统聚类分析、主成分分析和判别分析。结果发现,对于~1H-NMR波谱数据的聚类分析和主成分分析,能够有效鉴别出猪皮胶和工业用骨胶;Fisher判别分析能进一步区分驴皮胶和非驴动物皮胶样品,判别正确率达91.67%,表明核磁代谢组学方法适于进行阿胶及其制品的原料来源鉴别,值得在扩大样本深入验证的基础上进行推广应用。The origin identification is of great importance for the quality control of Colla Corii Asini and its products. In the present study, high-throughput and non-destructive ~1H nuclear magnetic resonance spectroscopy(~1H-NMR) technique was combined with multivariate statistical analysis methods to indentificate the origin of Colla Corii Asini from the perspective of metabolomics. Fourteen animal gelatin samples were collected for ~1H-NMR spectral assay, and the data sets reflecting the metabolomics characteristics of the samples were obtained after normalization under the same conditions. Then hierarchical cluster analysis(HCA), principal component analysis(PCA), and Fisher discriminant analysis(FDA) were conducted for data mining based on these ~1H-NMR spectral data sets. The results indicated that HCA and PCA could effectively identify the samples from pig skin or animal bones. FDA could further distinguish the donkey skin samples from those non-donkey skin ones with a correct discrimination rate of 91.67%, suggesting that the present method might provide an effective alternative for the origin identification of Colla Corii Asini and its products, and it was worth in-depth verification with enlarging samples for real application.
