摘 要:目的:对药典法和樟帮法枳壳生品饮片挥发油的急性毒性及化学成分进行比较研究。方法:将两种挥发油系列乳剂分别一次性灌胃给予小鼠,观察给药后小鼠的毒性反应和死亡情况,采用改良寇氏法计算两种挥发油对小鼠的半数致死量(LD_(50));采用GC-MS法对两种挥发油化学成分进行分析。结果:药典法枳壳生品和樟帮法枳壳生品的挥发油LD_(50)分别为4 234.38、6 478.46mg/kg;药典法枳壳生品挥发油鉴定出36个成分,占总量的99.05%,而樟帮法枳壳生品挥发油鉴定出23个成分,占总量的99.52%,两者共有19个成分。结论:根据毒性分级,药典法枳壳生品挥发油为低毒,而樟帮法枳壳生品挥发油为无毒。两种挥发油化学成分种类及含量的差异,可能是造成其毒性差异的原因。Objective:To compare the acute toxicity and chemical constituent of volatile oil of Fructus aurantii pieces made by the methods of Pharmacopoeia and Zhang-bang.Methods:The two volatile oil emulsionwere respectivelygavage to mice.The toxic reaction and death of micewere observed,and the mice median lethal dose(LD_(50)) was calculate by the modified Karber method.The chemical constituents of the volatile oil were analyzed by GC-MS.Results:The LD_(50) of volatile oil from Fructus aurantii pieces made by Pharmacopoeia method was 4 234.38 mg/kg,while the LD_(50) of volatile oil made by Zhang-bang method was 6 478.46 mg/kg.The volatile oil of Fructus Aurantii made by Pharmacopoeia method was identified 36 components,accounting for 99.05% of the total,while 23 components were identified made by Zhang-bang method,accounting for 99.52% of the total,andthere weresame 19 components in total.Conclusion:According to the toxicity classification,the volatile oil of Fructus Aurantii from Pharmacopoeia is low toxicity,while the volatile oil of Fructus Aurantii from Zhang-bang method is non-toxic.Differences between the two species and the chemical composition andcontents of volatile oil may be the cause of its toxicity differences.
