摘  要:目的:探讨不同剂量远志皂苷对D-半乳糖致衰老小鼠的抗衰老作用,及对环磷酰胺所致免疫低下小鼠免疫功能的调节作用。方法:将小鼠随机分为空白组、模型组、阳性组及远志皂苷高、中、低剂量组。实验一:除空白组外,其余各组均注射D-半乳糖溶液1.25g/kg建立衰老小鼠模型,高、中、低剂量组依次喂饲400、200、100mg/kg的远志总皂苷,4周之后分别测定各组小鼠的脾脏指数、胸腺指数、血清中过氧化氢酶(CAT)和超氧化物歧化酶(SOD)的活力和脑组织丙二醛(MDA)含量、SOD活力。实验二:除空白组外,其余各组腹腔注射环磷酰胺(30mg/kg)制作免疫抑制小鼠模型,空白组注射同等剂量的0.9%氯化钠溶液;远志皂苷高、中、低剂量组分别灌胃400、200、100mg/kg的皂苷,其余各组灌胃等量的蒸馏水。连续给药2周后,测量各组小鼠的脾脏指数和胸腺指数,并用ELISA法验测出血清细胞因子白介素2(IL-2)和白介素6(IL-6)的含量。结果:抗衰老作用研究表明,与模型组比较,高、中剂量组的胸腺指数和脾脏指数均显著降低(P<0.01,P<0.05),血清的CAT活力极显著升高(P<0.01);低剂量组的SOD活力显著升高(P<0.05)。高、中、低剂量组脑组织的MDA水平、SOD活力亦统计学差异(P<0.05)。免疫功能调节作用研究表明,与模型组比较,远志皂苷高、中剂量组均显著升高脾脏指数和胸腺指数(P<0.05,P<0.01);皂苷高剂量组的血清细胞因子IL-2和IL-6的含量均显著升高(P<0.01)。结论:远志皂苷具有抗衰老及增强免疫功能的作用。Objective:To discuss the anti-aging effect of different dose Polygala saponins on D-galactose induced aging mice,and the immune function regulation effect of it on cyclophosphamide induced immunosuppressed mice.Methods:Mice were randomly divided into six groups:blank group,model group,positive group,and high,medium and low dose Polygala saponin dose group.Except the blank group,other groups were injected with 1.25 g/kg D-galactose to build the aging mice model.High,medium and low dose group were successively fed with 400,200,100 mg/kg of total saponins in Radix Polygalae.After 4 weeks,the spleen index,the thymus gland index,serum catalase(CAT),the activity of superoxide dismutase(SOD) and the content of malondialdehyde(MDA) in brain tissue were detected.Except the blank group,other groups were injected with 30 mg/kg cyclophosphamide to build immunosuppressive mice model,while the blank group was injected with same dose normal saline.High,medium and low dose group were given a gavage of 400,200,100 mg/kg saponin,and other groups were given a gavage of distilled water of the same volume.After treatment for 2 weeks,spleen index and thymus index of each group were detected,and the content of the serum cytokines interleukin 2(IL-2) and interleukin 6(IL-6) were measured by ELISA method.Results:The research on anti-aging effect showed that,compared with the model group,the thymus index and spleen index of high and medium dose groups decreased significantly(P〈0.01,P〈0.05); CAT activity of serum increased significantly(P〈0.01); the SOD activity in low dose group increased significantly(P〈0.05).In the high,medium,and low dose group,the differences of the level of MDA and the SOD activity in brain tissue between every two groups were statistically significant(P〈0.05).The study on immune function regulation showed that,compared with the model group,the spleen index and thymus index in high and medium dose group were significantly increased(P〈0.05,P〈0.01),and the c
