止痛润肠浓煎饮防治混合痔术后便秘实验研究

摘　　要：目的:探讨止痛润肠浓煎饮防治混合痔术后便秘的机制。方法:将48只SD大鼠随机分为:A组(正常组)、B组(模型组)、C组(麻仁软胶囊组)和D、E、F组(止痛润肠浓煎饮低、中、高剂量组)。治疗后测算小肠活性炭凝胶推进长度和推进率;检测血清前列腺素E2(PGE2)水平;RT-PCR检测结肠水通道蛋白3(AQP3)m RNA表达水平;免疫组化检测结肠黏膜AQP3蛋白表达情况。结果:与B组比较,各治疗组小肠活性炭凝胶推进长度和推进率均提高(P<0.01);E、F组PGE2水平下降(P<0.01);C、D、E、F组AQP3 m RNA表达水平均上调(P<0.01);E、F组AQP3蛋白表达上调(P<0.01)。结论:止痛润肠浓煎饮可能通过促进肠蠕动、减轻肛门疼痛、调节AQP3表达等防治混合痔术后便秘。Objective：The purpose of this study is to research the mechanism of Zhitong Runchang Condensed Decoction in prevention and treatment of constipation after operation of mixed hemorrhoids.Methods：Forty-eight SD rats were randomized into group A（normal group）,group B（model group）,group C（Maren Soft Gelatin Capsule group） and group D,E,F（low,middle,high dose of Zhitong Runchang Condensed Decoction group）.After the treatment,the length of small intestinal transiting activated carbon gel was measured,and the rate of transiting was calculated.The level of serum prostaglandin E2（PGE2） was tested by ELISA.Aquaporin 3（AQP3） m RNA in colon was tested by RT-PCR.The expression of AQP3 in colon was detected by immunohistochemistry.Results：Compared with group B,the length and rate of small intestinal transiting activated carbon gel in therapy groups were all improved（P〈0.01）; The levels of serum PGE2 in group E and F were decreased（P〈0.01）; the levels of colon AQP3 m RNA expression in Group C,D,E,F were increased,the levels of the expression of AQP3 protein in group E,F were increased（P〈0.01）.Conclusion：Zhitong Runchang Condensed Decoction can prevent and treat constipation after mixed hemorrhoid by promoting intestinal peristalsis,alleviating anal pain and regulating AQP3 expression.
