摘 要:为了对陕西省羊血液样品中的泰勒虫(Theileria sp.)进行种类鉴定和遗传学分析,本试验利用形态学方法对采集的血液样品通过涂片和姬姆萨染色后进行显微镜观察,发现红细胞内存在大小1.0~1.5μm、原生质呈淡蓝色、染色质呈红色并处于红细胞内侧边缘的多形性虫体。利用分子生物学方法,对梨形虫主要表面蛋白(major piro—plasm surface protein,MPSP)基因进行PCR扩增、序列分析以及进化树构建,发现有2份DNA样品(TL001和TL002)扩增出大小约800bp的目的条带。序列比对和种系发育树表明,分离到的2个虫株与GenBank中neileria luwenshuni(GQ281044)序列相似性分别为100%和99%,且位于同一进化支,提示本试验所分离到羊血液原虫为吕氏泰勒虫(T.luwenshuni),同时也为该地区羊泰勒虫病的防制奠定基础。In order to conduct the species identification and phylogenetic analysis for Theileria sp. in infected goats in Shaanxi province, the microscopic observation of the blood smears subjected to Giemsa staining found the presence of polymorphic parasites with the size of 1.0-1. 5μm, light blue plasmids and red nucleus located in the edge of the erythrocytes. Moreover,the molecular bio- logical results indicated that two PCR products of DNA preparations(TL001, TL002) generated 800 bp target bands by agarose gel electrophoresis. Further study by the sequence alignment and the pbylogenetic tree found that the sequence similarities between the two isolates and T. luwen- shuni(GQ281044) were 100% and 99%, respectively. The result of the phylogenetic tree also showed that TL001,TL002 and T. luwenshuni(GQ281044) were grouped in a one clade,hinting a close phylogenetic relationship among them. Therefore, the blood protozoan isolated here was T. luwenshuni and the results of this study will provide some fundamental data for the preventing and controlling the theileriosis of goats and sheep in this region.
