摘 要:为研究新疆不同地区牛支原体分离株oppD/F基因的遗传特性,从哈密、塔城等新疆不同地区的规模化奶牛场采集了疑似牛支原体感染的病死奶牛肺脏组织、关节液及鼻拭子。对病料进行病原分离,PCR检测特异性基因op—pD/F及序列分析。结果显示,固体培养基上得到典型的“煎蛋状”菌落,并扩增出448bp特异性目的条带。生物信息学分析显示,24株新疆不同地区分离株间核苷酸和氨基酸序列同源性均较高,与国际标准株PG45(CP002188.1)氨基酸序列同源性为97.3%~100.0%,具有高度同源性。研究结果为新疆地区牛支原体流行病学分子信息及免疫预防提供理论依据。In order to study the genetic characteristics of oppD/F gene of Mycoplasma boris isola- ted from different regions of Xinjiang,this study collected lung tissue,joint fluid and nasal swab of cases of suspected Mycoplasma boris infection from the scale of dairy farms of Hami,Tacheng and other regions of Xinjiang,to carry out isolation of pathogen from the disease,PCR detection of spe- cific gene oppD/F and sequence analysis. Results showed that colonies of 24 strains isolated from different regions of Xinjiang province were typical fried-eggs on solid medium; and an expected gene fragment of 448 bp was amplified by PCR. Bioinformatics analysis displayed that the nucleo- tide and amino acid homology of 24 isolates were higher,the amino acids of sequnces had 97.3%- 100.0% of similarity to international standard strain PG45(CP002188.1). The results of the study provide a theoretical basis for the molecular information and immune prevention of Mycoplasma boris in Xinjiang.
