摘 要:目的探讨脂联素基因多态性与儿童单纯性肥胖的关联性。方法采用整群抽样方法,从内蒙古包头市7~13岁的蒙古族儿童中选取232例,其中116例肥胖儿童为肥胖组,116例健康儿童为正常体重组。检测两组对象的生化指标,利用聚合酶链反应-限制性片断长度多态性方法检测APN SNP+45(T/G)位点多态性。结果 (1)肥胖组和正常体重组FPG、TG、LDL-C和HDL-C水平差异均有统计学意义(P<0.05)。(2)两组间APN SNP+45(T/G)位点GG型、TG型、TT型基因型频率分别为13.79%、56.03%、30.17%和6.90%、42.24%、50.86%,两组基因型频率与等位基因频率分布差异有统计学意义(P<0.05)(3)肥胖组APN SNP+45(T/G)位点的TG+GG基因型与TT型相比,其TC、TG、LDL-C和FPG水平均升高,差异均有统计学意义(P<0.05);正常体重组中,TG+GG基因型与TT相比,TC、TG、HDL-C、LDL-C和FPG差异均没有统计学意义(P>0.05)。结论 APN SNP+45(T/G)多态位点与包头市蒙古族儿童单纯性肥胖的发生可能相关。Objective To investigate the association between adiponectin gene polymorphisms and simple obesity in children. Methods A total of 232 Mongolian children aged 7 - 13 years old in Baotou, Inner Mongolia were selected by cluster sampling. Among them, 116 obese children were included in the obesity group, while 116 healthy children were included in the normal weight group. The biochemical markers of the two groups were detected and the polymorphism of APN SNP + 45 (T/G) locus was detected by Polymerase chain reaction restrictive fragment length polymorphism method. Results There were significant differences between the levels of FPG, TG, LDL - C and HDL - C between normal and obesity group (P 〈 0.05). The GG,TG and TT genotype frequencies of APN SNP +45 (T/G)in obesity group of Mongolian children were 13.79% ,56.03% and 30.17%, and 6.90,42.24% and 50. 86% in the normal group. There was a significant difference between genotype frequency and allele frequency distribution in the two groups ( P 〈 0.05 ). At the APN SNP + 45 (T/G) locus, the TC, TG, LDL - C and FPG levels were significantly higher in obese children with TG + GG genotype than those in obese children with TT genotype, and the differences were statistically significant( P 〈 0.05 ). However, there was no significant difference in TC, TG, HDL - C, LDL - C and FPG levels between APN SNP + 45 (T/G) TG + GG genotype individuals and the ones with IT genotype in normal group (P 〉 0.05 ). Conclusion The polymorphisms of APN SNP + 45 (T/G) may be associated with the occurrence of simple obesity in Mongolian children in Baotou City.
