摘 要:传统的毛细管电色谱柱一般在检测窗口的前端填有固定相,后端为空管。在该毛细管电色谱柱内电导、电渗流和电场强度等诸多参数在检测窗口前后不同,这为毛细管电色谱的理论研究带来不便。与此同时,在检测窗口的两端存在压差,更易产生气泡,致使中断电流和实验。为了解决上述问题,建立了3种方法制备具有改进柱型的毛细管电色谱整体柱,即检测窗口的两端均填有固定相,而检测窗口没有固定相,并与传统的毛细管电色谱柱进行了比较,在相同条件下,柱效和中性溶质保留因子在两类毛细管电色谱柱上不存在明显差异,具有改进柱型的毛细管电色谱柱消除了电渗流、电场强度以及电导在检测窗口两端的差异,从而易于上述参数的测量和抑制气泡的产生,提高运行稳定性。Generally there is stationary phase ahead of detection window,while no stationary phase after the detection window in a capillary electrochromatography column with conventional configuration. In this kind of column,many parameters such as conductance,electroosmotic flow and electric field strength are different before and after the detection window,which brings inconvenience to the theoretical research on capillary electrochromatography. Furthermore,there is a pressure difference at two ends of the detection window,which causes bubbles and interrupts current and experiment. In order to solve the above problems,three methods for preparation capillary electrochromatography column with improved configuration were developed,in which there are stationary phases at both ends of detection window while no stationary phase at the detection window. Compared with the capillary electrochromatography column with traditional configuration,the column efficiency and retention factors of neutral solutes do not exist significant differences under the same experimental conditions,indicating the existence of stationary phase after the detection window does not have influence on column efficiency. The capillary electrochromatography column with improved configuration eliminates the differences in conductance,electroosmotic flow and electric field strength at the ends of the detection window,thus making the measurement of these parameters more convenient and helpful to suppress the generation of bubbles.
