摘 要:目的:检测S1P1和AT1R蛋白及mRNA在人肝癌组织中的表达。方法:对18例人正常肝组织和23例人肝癌组织进行HE染色。采用免疫组化、Real-time PCR及Western blot方法检测人正常肝组织和人肝癌组织中S1P1及AT1R蛋白及mRNA表达。结果:S1P1及AT1R在人正常肝组织的细胞膜和细胞浆中可见弱的棕黄色颗粒沉着,肝癌组织的细胞膜和细胞浆中可见强的棕黄色颗粒沉着。与正常肝组织相比,肝癌组织中S1P1和AT1R蛋白及mRNA表达水平显著增高(P<0.05),两者呈现一致趋势。结论:S1P1及AT1R在人肝癌组织中的蛋白及mRNA表达水平呈一致升高的趋势,提示两种分子之间可能存在一定的相关性,为进一步研究其在肝癌发生发展中的关系提供了一丝新线索,也为临床上肝癌的诊断和防治提供一些新策略。Objective To investigate protein and mRNA expressions of SI PI and ATI R in human liver cancer tis-sues. Methods: HE staining of 18 normal liver tissues and 23 human liver cancer tissues was observed. Moreover,pro-tein and mRNA expressions of SI PI and AT1R in normal liver tissues and human liver cancer tissues were determined with SP immuohistochemical method,Real - time PCR and Western blot. Results: Protein and mRNA expressions of SI PI and ATI R in human liver cancer tissues was significantly stronger than that in normal liver tissues ( P 〈 0. 05 ), presenting a consistent trend. Conclusion : Protein and mRNA expressions of SI PI and AT1R in human liver cancer tissues are enhanced, suggesting that there may be a correlation between the two molecules, and providing a new clue for further study of their relationship in the development of liver cancer,finally providing some new strategies for theprevention and treatment of liver cancer.
