摘  要:目的 分离并纯化伏康树籽中的吲哚类生物碱。方法 先采用传统的柱层析法分离其醇提物,再采用半制备HPLC系统进行纯化,收集对应峰的流出液,检测产品的纯度,结合理化常数测定及波谱学技术确定其结构。结果 通过传统柱层析法分离得到生物碱类化合物柳叶水甘草碱(Ⅳ),经过半制备液相色谱法分离得到3-oxotabersoine(Ⅰ)、(-)-pachysiphine(Ⅱ)、(-)-lochnericine(Ⅲ)等生物碱单体。结论 除化合物Ⅳ外,其余3个化合物均为首次从该属植物中分离得到。利用半制备色谱技术和传统柱层析法结合,分离纯化伏康树籽中结构类似的吲哚生物碱,具有简便、快捷和周期短等优点。OBJECTIVE To separate and purify the indole alkaloids from the seeds of Voacanga africana. METHODS V. Africana alcohol extracts were treated by traditional column chromatography, following by semi -preparative chromatography systems. The eluent of corresponding peak was collected and the purity of each isolated compound was determined by HPLC. Their structures were elucidated on the basis of physicochemical properties and spectroscopic analysis. RESULTS Tabersonine ( IV ) was obtained by traditional column chromatography,while three monomeric indole alkaloids [ 3 - oxotabersoine ( I ), ( - ) - pachysiphine ( H ) ,and( - ) -lochnericine(m) ] were purified by semi -preparative high performance liquid chromatography. CONCLUSION The latter three alkaloids have been found from Voacanga genus for the first time. The combination of semi - preparative high perform- ance liquid chromatography and traditional column chromatography is proved to be a simple, fast and short - cycle method in the separation and purification of structurally similar indole alkaloids from seeds of V. Africana.
