摘  要:目的 采用高效液相色谱-串联质谱法测定小鼠血浆中的肌酐浓度。方法 采用Shisheido C18色谱柱(100 mm×4.6mm,5μm),流动相为甲醇-水-甲酸(30∶70∶0.05),流速0.2 mL·min^(-1),内标为吉西他滨。以多反应离子监测,采用正离子模式,电喷雾离子源,血浆样品经沉淀蛋白后进样。结果 肌酐血浆的线性范围为0.5~20μg·mL^(-1),批内及批间RSD均小于7%,方法回收率为97.67%~103.11%。结论 所用方法操作简单、结果准确、重复性好,可用于测定小鼠血浆中的肌酐浓度。OBJECTIVE To determine creatinine in rat plasma using LC - MS/MS method. METHODS The analytical column was Shisheido C18 ( 100 mm × 4.6 ram,5 μm). The mobile phase consisted of methanol - water - formic acid( 30 : 70 : 0.05 ) with flow rate of 0.2 mL min-1. The column temperature was set at 40 ~C. Gemcitabine was used as an internal standard. The samples were precipitated before injection. RESULTS The calibration curves had good ]inearity within a concentration range of 0.5 - 20 μg. mL- 1. The intra - day RSD and inter - day RSD were less than 7%. The average recoveries for creatinine plasma concentrations were 97.67% - 103.11%. CONCLUSION This method provides a sensitive, accurate, precise and reliable analytical procedure for deter- mining of creatinine in rat plasma.
