五福饮对大鼠退变椎间盘聚蛋白聚糖与Ⅱ型胶原表达的影响

摘　　要：目的观察五福饮对大鼠退变椎间盘聚蛋白聚糖与Ⅱ型胶原表达的影响。方法将48只SD大鼠分成空白对照组、模型组及五福饮组各16只,模型组及五福饮组大鼠应用细针纤维环穿刺结合肿瘤坏死因子-α椎间盘内注射进行造模,造模成功后3组大鼠分别给予生理盐水、生理盐水及五福饮灌胃治疗8周。采用Micro-CT检测脊柱退变情况,HE染色观察椎间盘退变特征,RT-PCR检测椎间盘聚蛋白聚糖与Ⅱ型胶原mRNA的表达。结果造模后,大鼠椎间盘出现明显退变特征,表明造模成功。灌服五福饮8周后,大鼠造模椎间隙相对高度及组织学退变均有改善。五福饮组大鼠与模型组大鼠相比较,椎间盘聚蛋白聚糖与Ⅱ型胶原mRNA表达均有明显提高(P<0.05)。结论五福饮可防治椎间盘退变,提高聚蛋白聚糖与Ⅱ型胶原表达可能是其作用机制。Objective： To explore the effects of WufuDeeoction on the expression of aggrecan and type [[ col- lagen in degenerative disc of rats. Methods： 48 SD rats were randomly divided into the control group,the model group and Wufu Decoction group, 16 c.ases in each. The model group and Wufu Decoction group used Fine needle fibrous ring puncture combined with TNF-α intradiscal injection to make models. After the success of modeling,3 groups were given normal saline,saline and WuJh Decoction garage for 8 weeks. Micro-CT was used to detect spinal degeneration,and HE staining was used to observe the characteristics of intervertebral disc degeneration. The mRNA expression of aggrecan and type 11 collagen in degenerative disc were tested with RT-PCR. Results： Afer modeling,there was a marked degeneration of the interve^ebral disc in rats,which showed that the model was successful. After 8 weeks＇ perfusion of wufu Decoction,the relative height of the intervertebral space and the histo!ogic degeneration were improved in the rat model. Compared Wufu Decoctiou group with the model group, the mRNA expression of aggrecan and type Ⅱ collagen increaged significantly（P〈 0.05）. Conclusion： Wufu De- coction can prevent intelwertebral disc degeneration,improve the expression of aggrecan and type Ⅱ collagen, which may be its mechanism.
