摘 要:目的建立积雪草苷血药浓度高效液相色谱法(HPLC)的测定方法。方法根据有效治疗剂量将SD大鼠分成低、中、高剂量组,分别尾静脉注射给药14、42、126 mg·kg^(-1),采用Welchrom-C18柱(250 mm×4.6 mm,5μm)色谱柱,流动相为∶水-乙腈(24∶76),监测波长为221 nm,流速1.0 mL·min^(-1),柱温28℃,葫芦素B为内标测定血药浓度。结果积雪草苷在0.7~150.0μg·mL^(-1)范围内线性系良好(R^2=0.999),提取回收率均大于90%,相对回收率均大于99%,日内精密度均小于4%,日间精密度均小于7%,室温放置24 h稳定性相对标准偏差(RSD)均小于5%,-20℃环境冻融稳定性RSD均小于10%。结论 HPLC法灵敏、准确、快速,可用于积雪草苷血药浓度的测定。Objective To establish a method for the determination of Asiaticoside level in rats plasma by high performance liquid chromatography(HPLC). Methods HPLC method was used to determine the plasma concentration of Asiaticoside in SD rats with single dose of Asiaticoside(14,42 and 126 mg·kg^(-1))via tail vein. Analytical column was Gemini C18( 4.6 mm× 150 mm,5 μm); the HPLC profile: mobile phase was acetonitrile-glacial acetic acid(24∶76) at a flow rate of 1.0 mL/min.Detection wavelength was set at 221 nm and temperature was kept at 28℃. Cucurbitacin B was applied as internal standard.Results Asiaticoside showed good linear relationships in ranges of 0.7~150 μg · mL^(-1)( R^2=0.999). Extraction recoveries were greater than 90%. The relative recoveries were greater than 99%.Intra-day and inter-day relative standard deviation(RSD)values were less than 4% and 7%, respectively. The stability of RSD at room temperature for 24 hours and freezethaw stability of -20℃ were less than 5% and 10%, respectively. Conclusion The HPLC method was sensitive, accurate and rapid, and could be used for the determination of Asiaticoside level in rat serum.
