摘 要:目的对前期分离鉴定的嗜肺军团菌5型(LP5)进行毒力分析。方法构建CD-5感染巨噬细胞模型,瑞士染色后,光镜下观察细胞变化,再行透射电镜进一步观察,开展末端脱氧核苷酸转移酶介导的dUTP缺口末端标记测定(TUNEL)实验,研究CD-5是否为毒力株。结果成功构建CD-5感染巨噬细胞模型,光镜观察CD-5能使细胞空泡变性、破裂。透射电镜显示,细胞出现线粒体聚集、胞质空泡化,胞核皱缩,染色质凝集于胞膜周边等凋亡表现,TUNEL实验也检测到DNA断裂的片段。结论分离获得的CD-5为毒力株,有可能对人体致病,应加强对环境中军团菌污染情况的监测和控制,避免军团菌病的爆发和流行。Objective To analyze the virulence of Legionella bacteria strain numbered CD-5 that proved to be Legionella pneumophila serotype 5(Lp5)by the methods of molecular biology and Phylogenetic in Department of Parasitology,West China School of Preclinical and Forensic Medicine,Sichuan University. Methods The model of macrophage infected by CD-5 strain was constructed. The changes of the infected cells were observed under optical microscope after Swiss dyeing and further observed by transmission electron microscopy. Cell death was detected by the terminal dexynucleotidyl transferase(TdT)-mediated dUTP nick end labeling(TUNEL)method. Results Successfully constructed the model of Macrophage infected by CD-5 strain was constructed. Cell vacuoles degeneration and cell rupture were observed under optical microscope. Transmission electron microscopy showed apoptosis features,such as mitochondria condensation,cytoplasmic vacuoles,nucleus shrinkage,and chromatin condensation. The TUNEL assay identified nuclear DNA fragmentation.Conclusion The isolated CD-5 strain was virulence strain which could be a pathogenic form for human body. So measures should be taken to strengthen supervision and control for Legionella contamination in environment to avoid the outbreak and prevalent of Legionellosis.
