摘 要:目的探讨镇喘保肺汤对肾气虚哮喘大鼠尿液代谢的影响,为肾气虚哮喘证候的诊断与治疗提供方法。方法将36只雄性Wistar大鼠随机分为正常对照组,肾气虚哮喘模型组及中药组,每组各12R。除正常组外,其余两组均使用卵白蛋白注射联合恐伤肾及游泳力竭法建立支气管哮喘肾气虚证模型。中药组在实验第15天开始给予镇喘保肺汤2.0g/kg灌胃,正常组和模型组给予等量生理盐水灌胃。连续干预15d后收集晨尿,通过LC—MS技术进行检测,采用PCA分析结合OPLS—DA分析并确定差异性表达代谢物。结果与正常对照组比较,模型组大鼠尿液中肾上腺酮、二十四碳六烯酸、胍丁胺、硬脂酸含量显著升高,前列腺素(19:0/0:0)、前列腺素(16:0/0:0)、尿可的松、脱氧胆酸、甘油二脂、皮质酮、I-尿胆素、脱氧皮质酮含量明显下降。与模型组比较,中药组大鼠尿液中脱氧皮质酮、蛋白酶抑制剂[0—20:0/22:4(7Z,10Z,13Z,16Z)]、21-羟基孕烯醇酮等含量显著升高,镁[0:0/22:6(4Z,7Z,10Z,13Z,16Z,19Z)/0:0]、二十碳五烯酸、肾上腺酮、溶血性磷脂酸[20:3(8Z,11Z,14Z)/0:0]、胍丁胺、二十二烷酸等明显下降。结论肾气虚哮喘证候的出现可能与脂类代谢及肾上腺糖皮质激素分泌异常有关,而镇喘保肺汤则可能通过改善脂类代谢的途径,调节肾上腺糖皮质激素分泌预防和治疗支气管哮喘肾气虚证。Objective To probe the effect of zhenchuanbaofei decoction on urine metabolism of kidney qi in asthmatic rats, and to provide methods for asthma syndromes of kidney deficiency and diagnosis treatment. Methods 36 male Wistar rats were divided into blank control group, kidney qi deficiency asthma group and Chinese Medicine rats urine group randomly, each group had 12rats. Except blank group, the rest two groups were used the egg albumin injection combined with kidney injury and swimrmng exhaustion to establish the model of bronchial asthma with kidney qi deficiency syndrome. In the 15th day of the experiment, start 2.0g/kg intragastric administration of zhenchuanbaofei decoction, same dosage of NS to blank group. After 15 days continuous intervention, urine samples were collected and detected by LC-MS technique. PCA analysis and OPLS-DA analysis were used to identify the differentially expressed metabolites. Results Compared with the control group, the content ofadrenosterone, twenty- four carbon acid etc. in the kidney qi deficiency asthma group increased significantly, and the content ofPG ( 19: 0/0:0 ) and deoxycorticosterone etc. decreased significantly. Compared with the group of asthma kidney qi deficiency, the contents of deoxycorticosterone, 21-hydroxypregnenolone etc. in the traditional Chinese medicine group were significantly increased, and the contents of eicosapentaenoic acid, adrenosterone, agmatine, and so on, decreased significantly in the rats of the model group. Conclusion The syndrome of kidney qi deficiency asthma is perhaps related to lipid metabolism and abnormal secretion of adrenal cortical hormone, while the zhenchuanbaofei decoction may prevent and cure bronchial asthma by improving the way of lipid metabolism and regulate the secretion of adrenal cortex hormone.
