摘 要:目的建立高效液相色谱法(HPLC)测定热炎宁颗粒中野黄芩苷含量的测定方法。方法色谱柱:Ultimate XB-C18柱(4.6×250 mm,5μm);以乙腈-0.02 mol/L磷酸二氢钾溶液(用三乙胺调节p H值至5.0)(14∶86)为流动相;检测波长:335 nm;柱温:30℃;流速:1.0 ml/min;进样量:20μl。结果野黄芩苷在7.57~30.28 mg/L的浓度范围内线性良好,平均加样回收率为100.11%,相对标准偏差为0.3%。结论该测定方法灵敏度高、准确、可靠、重现性良好,可作为热炎宁颗粒测定野黄芩苷的质量控制方法。Objective To establish an HPLC method for the determination of the content of scutellarin in Reyanning granules. Methods Ultimate XB-C18 column (250 × 4.6 mm,5 μm) was used. Acetonitrile and O. 02 mol/L monopotassium phosphate( adjust the pH to 5.0 with triethylamine) at a ration of 14:86 was taken as the mobile phase. The sample was determined by HPLC at the wavelength of 335nm and the temperature of column was 30℃. The flow rate was 1.0 ml/min. Sample size was 20μl each time. Results Scutellarin had a good linear relationship within the range of 7.57 - 30.28 mg/L. The average recovery rates were 100.11% ,and relative standard deviation was 0.3%. Conclusion The method has the characteristics of high sensitivity, accuracy, reliability, and good reproducibility, which can be used as a quantitative analysis method for scutellarin in Reyanning granules.
