摘  要:目的:观察白芍木瓜汤对碘乙酸诱导的大鼠骨性关节炎的药效作用,并初步探讨其可能机制。方法:取雄性SD大鼠50只,按体质量随机分为正常对照组、模型对照组、阳性对照组、白芍木瓜汤高、低剂量组,每组10只。除正常对照组外,其余各组大鼠双侧膝关节腔注射碘乙酸建立骨性关节炎模型。术后第2 d各组大鼠开始给予药物干预,连续6周,每周称量大鼠体质量,检测大鼠抓力。第6周末,各组大鼠麻醉,进行X-ray活体成像观察;心脏取血,分离血清,酶联免疫法检测各组大鼠血清中基质金属蛋白酶13(MMP-13)、软骨蛋白聚糖(Aggrecan)、II型胶原(Col II)的含量;截取膝关节,剔除皮肤与肌肉,EDTA脱钙后分别进行HE染色与SO染色,光镜下观察各组大鼠膝关节病理变化。结果:造模后6周,相比于正常对照组,模型组大鼠体质量、血清MMP-13、Col-II、Aggrean含量均出现显著性差异(P<0.01),膝关节影像学上表现出明显损伤,光镜下观察到明显的病理改变;给药6周后,高、低剂量的白芍木瓜汤对OA大鼠体质量、抓力的影响不明显,但相比于模型对照组,白芍木瓜高、低剂量组大鼠血清MMP-13含量均显著降低(P<0.01),Col-II、Aggrean含量均显著升高(P<0.01),膝关节影像学损伤与光镜下病理改变均得到明显改善。结论:白芍木瓜汤对碘乙酸诱导的大鼠骨性关节炎有良好的药效作用,其作用机制可能与抑制MMP-13产生,促进Aggrecan、Col-II合成有关,从而达到修复软骨有关。Objective: To observe the pharmacological actions of Baishao Mugua Tang on osteoarthritis rat model induced by iodoacetic acid and preliminary explore its possible mechanism. Methods: According to the body weight, 50 male SD rats were randomly divided into normal control group, model control group, positive control group, high and low dose of Baishao Mugua Tang , 10 rats in each group. Rats were injected iodoacetic acid in bilateral knee joint cavity to induce osteoarthritis model. After the second day of surgery, started to give drug intervention to each group rats for six weeks continuously, weighing rat body weight and making detection of holding power in rats every week. Rats were done anesthesia, and then to make X - ray living imaging; then tak- ing blood from the heart and separating serum, the contents of metalloproteinases 13 (MMP - 13 ) , cartilage pro- teoglycan (Aggrecan) , type II collagen (Col - II) in in each group were determined by Enzyme -linked immu- noassay. Making an interception of knee joint, removing the skin and muscle, after decalcified with EDTA, dye- ing HE and SO respectively, then observing the knee joint pathological changes under light microscopy in each group rats. Results: After modeling 6 weeks, compared with normal control group, body weight of model group rat, serum levels of MMP - 13, Col - II, Aggrean showed significant difference (P 〈0.01 ) , the knee joint im- aging showed obvious damage, the obvious pathological changes were observed under light microscopy. After drug delivery for 6 weeks, the changes of holding power and body weight of OA rats in high and low doses Baishao Mugua Tang groups were not obvious, but compared with model control group, serum MMP - 13 level of high and low dose group of rats were significantly lower (P 〈 0.01), Col- II, Aggrean contents were significantly increased (P 〈0. 01), both knee injury on imaging andpathological changes under light microscopy were improved significantly.
