摘 要:目的:观察三七多糖对糖尿病肾病大鼠的治疗效果及其对炎症反应和脂质代谢的调节作用。方法:60只大鼠随机分为正常组、模型组和三七多糖组,每组20只。模型组和三七多糖组给予高脂饲料喂养4周后,进行链脲佐菌素(STZ,50 mg/kg)注射造模,糖尿病肾病造模成功后,三七多糖组予以三七多糖(250 mg/kg·d-1)灌胃,正常组、模型组大鼠予以相同体积生理盐水灌胃,分别在6、12周时处死大鼠各半,采血测定血糖(BG)、胰岛素(INS)水平,计算胰岛素抵抗指数(HOMA-IR),并检测大鼠肾功能指标(BUN、SCr)及血脂水平,ELISA法测定血清炎性因子IL-1β、IL-6、TNF-α含量。取肾组织,HE染色,光镜观察病理变化,分别用RT-q PCR和Western blot方法进行SREBP-1c(固醇调节元件结合转录因子-1c)、ACCα(乙酰辅酶A羧化酶α)基因和蛋白表达的测定。结果:与正常组比较,模型组大鼠血清BG、INS、BUN、SCr、IL-1β、IL-6、TNF-α、TG、TC和LDL-C含量均明显升高(P<0.05),HOMA-IR明显升高;肾组织SREBP-1c、ACCαmRNA和蛋白表达明显增强(P<0.05)。三七多糖给药6周后,上述指标下降明显(P<0.05),且肾脏组织中SREBP-1c、ACCαmRNA和蛋白表达下调(P<0.05);给药12周后,上述指标改善更显著。结论:三七多糖可能通过减轻炎症、改善脂质代谢紊乱起到保护肾脏、改善DN的作用。Objective: To observe the treatment effect of Panax Polysaccharides on diabetic nephropathy (DN) rats and investigate the regulation effects on inflammation and dyslipidemia. Methods: 60 male SD rats were divided into normal control group , model group and Panax Polysaccharides group randomly with each 20 rats. The rats in model and Panax Polysaccharides groups were fed with a high - fat diet, 4 weeks later, STZ so- lution were given (50 mg/kg) by intraperitoneal injection , after the establishment of DN rats model, the rats of Panax Polysaccharides group were fed with Panax Polysaccharides (250 mg/kg · d- 1 ) , while the model group and control group received normal saline with the same method respectively once a day, 6 weeks and 12 weeks , half of the rats were sacrificed in each group, BG, INS, TC, TG, LDL- c levels were determined and HOMA -IR was calculated, serum IL - 1 β, IL - 6, and TNF - α were determined by ELISA, RT - qPCR and Western Blot were applied to detect renal SREBP - 1 c and ACCcmRNA and protein expressions. Results : Compared with the normal control group, BG, INS and serum BUN, SCr, IL - 1 β, IL - 6, TNF - co, TG, TC and LDL - C contents and HOMA - IR in rats of the model group were up - regulated, renal SREBP - 1 c and ACCcmRNA and protein expressions were increased significantly ; all these changes in Panax Polysaccharides group were down - regulated obviously after given panax polysaccharides for 6 weeks ( P 〈 0.05 ), the improvement of all above indexes were more significant after given Panax Polysaccharides for 12 weeks. Conclusion: Panax polysaccharides may protectkidney from injury, ameliorate the DN pathological changes by regulating the inflammatory reaction and dyslipidemia.
