摘 要:目的:观察益元生血方提取物对再生障碍性贫血(AA)患者T淋巴细胞亚群的影响。方法:收集AA患者10例为观察组,健康者10例为对照组。抽取受试者外周血制备淋巴细胞悬液,将益元生血方水提物用完全RPMI-1640稀释至以下浓度10%、5%、2.5%、1.25%、0.625%并干预AA患者淋巴细胞悬液,分别培养24、36、48、72小时后检测不同浓度、不同时间益元生血方水提物培养的T淋巴细胞增殖的活性,以确定益元生血方水提物作用的最佳药物浓度和最佳培养时间。流式细胞术检测2组外周血T淋巴细胞亚群变化。结果:当培养时间定为36小时,药物浓度定为5%时细胞增殖活性最佳。与正常对照组比较,观察组外周血CD4^+、CD4^+/CD8^+、白细胞介素4(IL-4)、CD4^+CD25^+的表达明显下降(P<0.05),CD8^+、干扰素γ(IFN-γ)、白细胞介素17(IL-17)的表达显著升高(P<0.05)。与干预前比较,干预后CD4^+、CD4^+/CD8^+、IL-4、CD4^+CD25^+的表达下降(P<0.05),CD8^+、IFN-γ、IL-17的表达显著升高(P<0.05)。结论:益元生血方可通过调节AA患者外周血T淋巴细胞亚群分化,调节免疫,从而发挥治疗作用。Objective: To observe the influence of the extracts from Yi Yuan Sheng Xue prescription on T lymphocyte subsets of the patients with aplastic anemia(AA). Methods: Ten AA patients were collected as the observation group, ten healthy persons as the control group. Peripheral blood was drawn from the subjects to prepare the lymphocyte suspension, and the suspension was intervened after diluting aqueous extract of Yi Yuan Sheng Xue prescription with complete R PMI-1640 to the concentrations of 10%, 5%, 2.5%, 1.25% and 0.625%, after the suspension was cultured for 24, 36, 48 and 72 hours respectively, the optimal drug concentrations and the optimal culture time of aqueous extract of Yi Yuan Sheng Xue prescription were ensured by detecting the activity of T lymphocyte proliferation cultured by aqueous extract of Yi Yuan Sheng Xue prescription in different concentrations for different time, the changes of T lymphocyte subsets were detected in both groups by flow cytometry. Results:When the culturing time was 36 hours, medicinal concentration was 5%, the activity of cellular proliferation was the best. Compared with normal control group, the expressions of CD4^+, CD4^+/CD8^+, IL-4, CD4^+CD25^+decreased notably in peripheral blood of the observation group(P <0.05), the expressions of CD8^+, IFN-γ and IL-17 raised significantly(P<0.05). Compared with before intervention, the expressions of CD4^+, CD4^+/CD8^+, IL-4, CD4^+CD25^+lowered(P<0.05), the expressions of CD8^+, IFN-γ and IL-17 raised significantly after the intervention(P<0.05).Conclusion: Yi Yuan Sheng Xue prescription could play the roles in the treatment by regulating T lymphocyte subsets differentiation in peripheral blood of AA patients and immunity.
