摘 要:目的建立并优化D101型大孔吸附树脂分离纯化栀子大黄汤有效部位群条件。方法采用紫外-可见分光光度法测定总环烯醚萜苷、总异黄酮、总黄酮、总蒽醌的含量,以总环烯醚萜苷、总异黄酮、总黄酮、总蒽醌的含量作为考察指标,分别考察了上样量、上样浓度、洗脱剂浓度等因素对分离纯化工艺的影响。结果最佳工艺条件为上样量:树脂量(g∶ml)=1∶5;上样浓度按原药材计为0.5g/ml;吸附时间为3 h;每个浓度的洗脱剂用量为6个柱体积,流速控制2个柱体积/h。30%乙醇、60%乙醇、70%乙醇可以分别将总环烯醚萜苷、总黄酮、总异黄酮和总蒽醌洗脱完全。结论该分离纯化工艺合理、稳定,可用于工业生产。Objective To isolate and purify the effective fractions of Zhi - Zi - Da - Huang Decoction, D - 101 macroporous resin was used to separate the components with the optimal conditions. Methods UV - Vis spectrophotometry was used to measure the total content of iridoid glycoside, flavone, isoflavone and anthraquinone contained in these effective fractions. The content of total iridoid glycoside, isoflavones, flavonoids and anthraquinone were taken as the targets to investigated the influence of the sample weight, sample concentration and the concentration of eluent on the separation and purification process. Results The optimal ratio of loading amount of extractive to resin concentration was 1:5 (g:ml). The optimum sample concentration was 0.5g/ml in terms of the raw materials. 30% ethanol, 60% ethanol and 70% ethanol could completely elute iridoid glycosides, flavonoids, isofla- vones and anthraquinone, respectively. Conclusion This optimized separation and purification technology was reasonable and sta- ble, and it could be extended to large -scale production applications.
