灵芝多糖可改善机体T细胞亚群及AQP1、AQP3水平

摘　　要：为了观察灵芝多糖对荷膀胱癌T24细胞裸鼠T细胞亚群及AQP1、AQP3水平的影响。本研究取2014年1月~2016年5月医院进行的裸鼠30只作为研究对象,建立荷膀胱癌模型。采用随机数字方法将裸鼠分为氯化钠溶液组（n=10）、顺铂组（n=10）及顺铂~＋灵芝多糖组（n=10）。3组分别给予氯化钠、顺铂、顺铂~＋灵芝多糖腹腔灌胃治疗,连续治疗18 d。采用流式细胞仪测定3组治疗前、后T细胞亚群;采用Western blotting法检测3组治疗前、后AQP1、AQP3水平。结果显示,倒置显微镜下可见原代细胞培养后紧贴在瓶底生长,细胞之间连接疏松、饱满透亮,细胞数量相对较少;第三代细胞经培养后会变大、变圆,均贴壁生长,细胞多为长梭形结构,细胞的数量较多。顺铂~＋灵芝多糖组治疗18 d后,CD3~＋、CD4~＋及CD3~＋/CD8~＋的水平显著高于顺铂组及氯化钠溶液组（p〈0.05）;顺铂组治疗18 d后,CD3~＋、CD4~＋及CD3~＋/CD8~＋的水平显著高于氯化钠溶液组（p〈0.05）。顺铂~＋灵芝多糖组治疗18 d后,AQP1、AQP3水平显著高于顺铂组和氯化钠溶液组（p〈0.05）;顺铂组治疗后18 d后,AQP1、AQP3的水平显著高于氯化钠溶液组（p〈0.05）。因此,本研究认为裸鼠荷膀胱癌给予腹腔灌注顺铂~＋灵芝多糖治疗,能改善机体T细胞亚群及AQP1、AQP3水平,为进一步的临床研究提供了一定的理论依据。The purpose of the study was to observe the effect of Ganoderma lucidum polysaccharides on T cell subsets and AQP1, AQP3 expression of bladder cancer T24 cell line bearing mice. 30 nude mice from January2014 to May 2015 were selected as bladder models. Moreover, they were divided into sodium chloride solution group, cisplatin group, and cisplatin plus Ganoderma lucidum polysaccharide group by random number method,which were given intraperitoneal perfusion of sodium chloride, cisplatin, cisplatin combined with ganoderma lucidum polysaccharides, respectively, for 18 successive days. T cell subsets in three groups were examined by flow cytometer before and after the treatment. AQP1, AQP3 levels were detected by Western blotting. The result showed the cultured primary cell was closely attached to the bottom of the bottle under inverted microscopy, the cells were loosely connected, full and transparent, and the number of cells was relatively small. After culture, the third generation cells became larger and round, all were adhered to the wall, and the cells were mostly long spindle where there were more cells. After the treatment for 18 d in cisplatin combined with Ganoderma lucidum polysaccharides group, the levels of CD3~＋, CD4~＋and the levels of CD3~＋/CD8~＋were significantly higher than those of sodium chloride solution group and cisplatin group. The levels of CD3~＋, CD4~＋and CD3~＋/CD8~＋in cisplatin group were significantly higher than those of the sodium chloride solution group（P〈0.05）. After the treatment for 18 d in cisplatin combined with Ganoderma lucidum polysaccharides group, the levels of AQP1, AQP3 were obviously higher than those of sodium chloride solution group and cisplatin group. The levels of AQP1, AQP3 in cisplatin group were significantly more than those of the sodium chloride solution group（P〈0.05）.
