寻常性银屑病患者外周血CD14^+单核细胞中白细胞介素-17的表达及意义

摘　　要：目的:探讨寻常性银屑病患者外周血CD14^+单核细胞中白细胞介素(IL)-17的表达及意义,以期为该病的临床治疗提供新的理论依据。方法:应用密度-梯度离心法分别获取寻常性银屑病患者及健康献血者(正常对照)的外周血CD14^+单核细胞,应用酶联吸附免疫(ELISA)法和逆转录聚合酶链反应(RT-PCR)法检测两组人群外周血CD14^+单核细胞培养上清液中IL-17含量及其基因表达;扫描电镜观察与寻常性银屑病患者血清共培养后正常人外周血CD14^+单核细胞的形态变化。结果:寻常性银屑病患者组外周血CD14^+单核细胞培养上清液中IL-17含量及IL-17m RNA相对表达量分别为(28.79±5.96)ng/L和(4.48±2.64)ng/L,均高于健康献血者,差异有统计学意义(P<0.05)。健康献血者的外周血CD14^+单核细胞与寻常性银屑病患者血清共培养后,随着共培养时间的延长,细胞膜发生明显改变,甚至破裂。结论:寻常性银屑病患者外周血CD14^+单核细胞过度表达IL-17,此细胞及其细胞因子在该疾病的发生、发展中起一定的促进作用。Objective： To investigate the expression and significance of the interleukin-17 secreted by the CD14＋ monocytes of the patients with psoriasis vulgaris in order to provide rationale for the treatment of psoriasis. Methods： CD14＋ monocytes were from peripheral blood of patients with psoriasis vulgaris and healthy controls. The protein and mRNA of IL-17 in the culture medium were measured, using ELISA and RT-PCR, respectively. At the same time, the cellular morphology of peripheral blood CD14＋ monocytes was observed using scanning electron microscope after co-cultured with the serum from patients with psoriasis vulgaris. Results： Both protein and mRNA levels of IL-17 in the culture medium of patients with psoriasis vulgaris were significantly higher than that in normal controls （28.79±5.96 ng/L for protein and 4.48±2.64 ng/L for RNA）. Addition of serum from patients with psoriasis vulgaris to CD14＋ monocytes from healthy donors changed morphology of cell membrane or induced cell rupture. Conclusions： The over-expression of IL-17 in CD14＋ monocytes of patients with psoriasis vulgaris may play roles in the pathogenesis of psoriasis.
