摘 要:目的:明确益气养阴清热化瘀方对ITP的治疗作用并观察该方对Notch信号分子的影响,探讨其发挥效应的潜在机制。方法:60只小鼠按随机数字表法分为正常组10只、ITP模型组50只,于模型组小鼠腹腔每日注射含2μg大鼠抗小鼠CD41单克隆抗体(MWReg30单抗)的磷酸盐缓冲液200μL,造成持续的血小板减少症,建立ITP小鼠模型。第8天进行模型评估后,将模型组随机分为不同的治疗组进行药物干预2周,治疗结束后测量各组血小板计数;采用RT-PCR、Western blot法分别测量各组骨髓单个核细胞(BMMNC)Notch受体Notch1-4和Notch配体Jagged1,Jagged2基因和蛋白的表达。结果:与模型组比较,中药各剂量组均能显著升高血小板水平,明显降低Notch(1-4)、Jagged1、2基因和蛋白的表达。结论:益气养阴清热化瘀方能够通过下调Notch信号的表达水平对ITP的病理环节产生影响,从而发挥升板作用,达到治疗ITP的目的。Objective: To clarify the effect of Yiqi Yangyin Qingre Huayu Decoction for ITP and observe the influence for Notch molecular,exploring the potential mechanism for the treatment of ITP. Methods: 60 mice randomly divided into normal group( n = 10) and ITP model group( n = 50),BALB/C mice were rendered thrombocytopenia by daily intraperitoneal injection of 2 μg rat anti-mouse CD41 monoclonal antibodies( MWReg30) in 200 μL phosphate-buffered saline( PBS) in model group. On the 8 th day,After the evaluation,the ITP model mice were divided into different drug intervention groups which was performed with different drug interventions for 2 weeks,the platelet count was observed after treatment and then the gene and protein expression levels of Notch1-4 and Jagged1-2 in the bone marrow mononuclear cells( BMMNC) were measured by RT-PCR and Western blot respectively. Results: Each dose of Chinese medicine treatment group can significantly increase the platelet compared to model group and reduce notably Notch( 1-4),Jagged1,2 gene and protein expression levels. Conclusion: Yiqi Yangyin Qingre Huayu Decoction plays a role in elevating platelet level by reducing the expression of Notch signaling to influence the pathology progress of ITP and achieves eventually the purpose of treatment of ITP.
