摘  要:目的:观察黄连素对2型糖尿病(T2DM)伴非酒精性脂肪肝(NAFLD)大鼠肝脏的治疗作用,并探讨其作用机制。方法:采用高脂饲料合并小剂量链脲佐菌素(STZ)建立T2DM伴NAFLD模型,将其随机分为模型组和黄连素组,同性别、周龄的未造模动物作为正常组。灌胃给药20周,检测肝功、血脂及肝组织SOD、CAT、GPX、TC和TG含量。肝组织切片染色观察其病理改变。免疫组化检测FAS表达。Q-PCR检测CAT、GPX1及FAS的m RNA表达。结果 :黄连素可显著降低模型大鼠血清中ALT、AST、TC、TG和LDL-C水平及肝组织中的TC、TG含量,升高肝脏SOD、CAT、GPX活性,改善了模型大鼠的肝细胞脂肪变性及炎细胞浸润。黄连素能显著抑制FAS蛋白表达,上调肝脏中CAT、GPX1的m RNA水平,降低FAS的m RNA表达。结论 :黄连素对T2DM伴NAFLD大鼠的肝脏具有保护作用,其机制与改善脂质代谢、增强抗氧化酶活性有关。Objective:To observe the therapeutic effect of berberine on the liver of type 2 diabetes mellitus(T2DM)with non-alcoholic fatty liver(NAFLD),and to explore its mechanism.Methods:High fat feed stuffmerge small doses of chain urea with cephalosporins(STZ)T2DM with NAFLD rat model was set up.These rats were randomly divided into model group and Berberine group,other unmolded rats with the same weeks of age and gender as normal group.After management for 20 weeks,the serum ALT,AST,TC,TG and LDL-C levels were detected,and the liver tissue SOD,CAT,GPX,TC and TG were tested.Histopathological changes were observed in liver biopsy staining.Immunohistochemistry for FAS expression and Q-PCR for m RNA expression of CAT,GPX1 and FAS were detected.Results:Berberine could significantly reduce the levels of serum ALT,AST,TC,TG and LDL-C;the content of TC and TG in liver tissue of rats in Berberine group.It could also elevate the liver SOD,CAT and GPX activities of rats in Berberine group,and downgrade their liver sternosis and inflammatory cells infiltration.Further studies showed that berberine significantly inhibited the expression of FAS protein,increased m RNA levels of CAT and GPX1 in the liver,and reduced m RNA expression of FAS.Conclusion:Berberine can protect the liver of T2 DM rats with NAFLD,and its mechanism is related to the improvement of lipid metabolism and antioxidant enzyme activity.
