肾络宁干预糖尿病肾病大鼠病变进展研究

摘　　要：目的:观察肾络宁对实验性糖尿病肾病大鼠肾脏结构和功能的影响。方法:采用腹腔注射链脲佐菌素(STZ)联合高脂高糖饲料喂养建立糖尿病肾病(DN)大鼠动物模型,进行为期6周的药物干预。观察实验各组DN模型大鼠一般状态、血清总蛋白(TP)、血清白蛋白(ALB)、血清尿素氮(BUN)、血清肌酐(Scr)、血脂(TG)、血清总胆固醇(TC)和尿微量白蛋白等参数的改变;并观察各组肾组织形态结构变化。结果:经治疗6周后,西药组和中药组DN大鼠各项实验参数变化较模型组均有不同程度的改善,具有显著性差异(P<0.05);肾脏结构改变方面,西药组、中药组DN大鼠肾脏病理形态学改变较模型组受损程度均有所减轻。结论:初步证实肾络宁可有效改善糖尿病肾病大鼠的肾脏功能及减轻肾脏病理损害的作用,延缓疾病进展,但其作用机制有待进一步明确。Objective ： To observe the effect of Shenluoning on structure and function of kidney in experimental rats with diabetic nephropathy. Methods： Animal models of rats with diabetic nephropathy （DN） were established by intraperitoneal injection of streptozotocin （STZ） and feeding of high fat and sucrose diet. The models were intervened with Shenluoning drug for 6 weeks. In each experimental group of DN model rats ,the general state and the changes of serum total protein （TP）, serum albumin （ALB）, serum urea nitrogen （BUN）, serum creatinine （Scr）, serum total cholesterol （TC） and urinary microalbumin etc. parameters were observed. And the changes of from and structure of kidney tissu were observed in each group. Results ： After 6 weeks of treatment, the changes of various experimental indexes of DN rats in western medicine group and traditional Chinese medicine （TCM） group were all improved to some extent compared with model group, and with significant difference （ P 〈 0. 05 ） ; in structural change of kidney, compared with the damage degree of model group, the change of kidney pathomorphology of DN rats in western medicine group and TCM group was lessened. Conclusion： It is confirmed prelimina- rily that Shenluoning can effectively improve the renal function and renal pathological damage of DN rats, and delay the progress of disease, but its mechanism of action needs to be clarified further.
