摘 要:目的分析糖尿病大血管病变大鼠血管内皮生长因子(VEGF)、脂联素(APN)表达及生化指标检测结果,并分析其之间相互调节机制。方法选取24只正常健康SD雄性大鼠,随机分为正常对照组和糖尿病组,各12只。正常组正常喂养,糖尿病组制作糖尿病大血管病变模型。28周后,对比2组体质量、血糖和血脂检测结果,并采用酶联免疫吸附双抗体夹心法测定血清VEGF和APN水平,对比组间差异,并应用免疫组化法测定VEGF及APN在血管内皮细胞的表达。结果1、糖尿病组血糖、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)水平均高于正常组,且前者体质量低于后者,组间对比均发现有统计学差异(P<0.05),而2组胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)水平比较差异均无统计学意义(P>0.05);2、糖尿病组血清VEGF高于正常组,且前者APN水平远低于后者,均存在统计学差异(P<0.05);3、免疫组化显示:糖尿病大鼠组血管内皮细胞核VEGF呈强阳性表达,而正常对照组未见此改变。而正常对照组血管内皮细胞核中APN呈强阳性表达,而糖尿病大鼠组未见此改变。4、脂联素浓度和血管内皮生长因子浓度呈明显负相关(r=-0.845,P<0.05)。结论糖尿病可影响体质量、糖脂代谢,且血清VEGF水平异常升高,APN水平异常降低,推测脂联素浓度和血管内皮生长因子浓度呈明显负相关。Objective To analyze the expression of vascular endothelial growth factor(VEGF), adiponectin(APN) and biochemical indexes in diabetic rats with macrovascular disease, and analyze the mutual regulation mechanism between them. Methods 24 healthy male SD rats were randomly divided into normal control group and diabetic group, 12 rats in each group. Normal feeding, diabetic group made diabetic macroangiopathy model.After 28 weeks, compared two groups of body weight, blood glucose and blood lipid test results, and the determination of serum VEGF and APN levels of the enzyme by ELISA double antibody sandwich method, the differences between the two groups, the expression and immunohistochemical determination of VEGF and APN in vascular endothelial cells were made. Results 1.In diabetes mellitus group, blood glucose,triglyceride(TG) and low density lipoprotein cholesterol(LDL-C) levels were higher than that of the normal group, and the weight was lower than that of the latter, the comparison between the two groups were found to have statistically significant difference(P<0.05), while the cholesterol(TC) and high density lipoprotein cholesterol(HDL-C) level showed no significant difference(P>0.05); 2.The serum VEGF in diabetic group was higher than that in normal group, and the level of APN in the former was much lower than that in the latter group,and there was statistical difference(P<0.05); 3.Immunohistochemistry showed that vascular endothelial cells in group VEGF diabetic rats showed strong positive expression, and the normal control group was normal. The changes of vascular endothelial cells in APN had positive expression, and no diabetic rats group had this change. 4.The concentration of adiponectin was negatively correlated with the concentration of VEGF(r=-0.845,P<0.05). Conclusion Diabetes mellitus can affect body weight, glucose and lipid metabolism, and abnormal increase of serum VEGF level and abnormal decrease of APN level.
