摘  要:目的:探讨五虎汤对病毒诱发幼年SD哮喘大鼠肺组织初始CD4+T细胞转录因子mRNA及蛋白表达的影响。方法:采用RSV诱导,OVA致敏加激发制备大鼠哮喘模型。造模成功后,随机分成五虎汤高、中、低(4.752g/kg、2.376g/kg、1.188g/kg)剂量组,阳性对照组(地塞米松片,2.4mg/kg),哮喘模型组和空白对照组6组。治疗14d后,取肺组织,分别采用RT-PCR及免疫印迹法检测肺组织初始CD4+T细胞转录因子T-bet、GATA-3、RORγt、Foxp3 mRNA及其蛋白表达情况。结果:模型组T-bet、Foxp3 mRNA及蛋白较正常对照组表达降低(P<0.05),GATA-3、RORγt mRNA及蛋白较正常对照组表达升高(P<0.05);与模型组比较,五虎汤干预后,高剂量组T-bet、Foxp3 mRNA及蛋白表达显著升高(P<0.01),GATA-3、RORγt mRNA及蛋白表达显著降低(P<0.01);中剂量组T-bet、Foxp3 mRNA及蛋白表达升高(P<0.05)、GATA-3、RORγt mRNA及蛋白表达降低(P<0.05)。结论:五虎汤通过抑制转录因子GATA-3、RORγt表达、促进转录因子T-bet、Foxp3表达,进一步调控Thl/Th2、Th17/Treg细胞平衡和免疫功能进而抑制哮喘气道炎症形成。Objective: To investigate the effect of Wuhu decoction on the mRNA and protein expression of transcription factors in naive CD4^+T cells in lung tissue in young Sprague-Dawley rats with virus-induced asthma. Methods: A rat model of asthma was established by RSV induction and OVA sensitization and challenge. After the model was successfully established,the rats were randomly divided into low-,medium-,and high-dose Wuhu decoction groups( 4. 752,2. 376,and 1. 188 g/kg,respectively),positive control group( dexamethasone tablets,2. 4 mg/kg),asthma model group,and blank control group. After 14 days of treatment,lung tissue was collected and frozen. RT-PCR and Western blotting were used to measure the mRNA and protein expression of transcription factors T-bet,GATA-3,RORγt,and Foxp3 in naive CD4^+T cells in lung tissue. Results: Compared with the normal control group,the model group had significant reductions in the mRNA and protein expression of T-bet and Foxp3 and significant increases in the mRNA and protein expression of GATA-3 and RORγt( all P〈0. 05). Compared with the model group,the high-dose Wuhu decoction group had significant increases in the mRNA and protein expression of T-bet and Foxp3 and significant reductions in the mRNA and protein expression of GATA-3 and RORγt( all P〈0. 01),and the medium-dose Wuhu decoction group also had significant increases in the mRNA and protein expression of T-bet and Foxp3 and significant reductions in the mRNA and protein expression of GATA-3 and RORγt( all P〈0. 05). Conclusion: By inhibiting the expression of GATA-3 and RORγt and promoting the expression of T-bet and Foxp3,Wuhu decoction further regulates the balance of Th1/Th2 and Th17/Treg cells and immune function and thus inhibits the development of airway inflammation induced by asthma.
