摘 要:目的:观察跌打促愈片对家兔骨折愈合过程中VEGF(血管内皮细胞生长因子)微环境的影响,从微环境探索跌打促愈片促进骨折愈合的间接机制。方法:普通级新西兰大白兔36只,骨缺损模型复制成功后,将实验动物随机分为3组,即A组:模型组,B组:实验组(跌打促愈片组),C组:实验组+Rhu Mab VEGF组(VEGF拮抗剂)。光镜下观察骨细胞标本并用酶联免疫吸附法(ELISA)检测血清VEGF浓度。结果:光镜下B组骨折断端毛细血管生长、骨痂愈合情况、骨细胞活跃程度、正常骨组织形成时间等均优于A组,C组相对B组在骨折初期坏死骨细胞增多,新生毛细血管不足,骨折断端由纤维结缔组织充填。血清VEGF浓度检测结果显示B组明显高于A组及C组,组间比较差异有统计学意义(P<0.05)。结论:跌打促愈片通过改善骨折愈合部位的微环境,促进VEGF的表达,从而介导骨折愈合。Objective: To investigate the effect of Dieda Cuyu tablets on the microenvironment of vascular endothelial growth factor( VEGF) during fracture healing in rabbits,as well as the indirect mechanism of action of Dieda Cuyu tablets in promoting fracture healing from the aspect of microenvironment. Methods: A total of 36 conventional New Zealand white rabbits were selected. After the establishment of the model of bone defect,these rabbits were randomly divided into model group( group A),experimental group( Dieda Cuyu tablets,group B),and Dieda Cuyu tablets + Rhu Mab VEGF group( VEGF antagonist,group C). Bone cell samples were observed under a light microscope,and ELISA was used to measure the serum level of VEGF. Results: Under the light microscope,group B had better results in growth of capillaries at the fractured end,callus healing,bone cell activity,and time to the formation of normal bone tissue than group A; compared with group B,group C had an increase in necrotic bone cells and a lack of new capillaries in the incipient stage of fracture,and the fractured end was filled by fibrous connective tissue. Group B had a significantly higher serum level of VEGF than groups A and C( P〈0. 05). Conclusion: Dieda Cuyu tablets can improve the microenvironment of fracture healing site,promote the expression of VEGF,and thus mediate fracture healing.
