摘 要:通过生物信息学方法从番木瓜基因组中获得一个泛素活化酶基因CpUBA1。利用软件分析该基因的序列特性,同时利用荧光定量PCR技术分析CpUBA1基因在番木瓜果实成熟过程中的表达模式,结果表明,CpUBA1基因序列开放阅读框为3 459 bp,编码的蛋白具有1 152个氨基酸,其分子量为128.93 ku,理论等电点为5.49,定位于细胞核。荧光定量PCR分析发现,CpUBA1基因在番木瓜叶片中的表达量最高,在雌花中的表达量最低。CpUBA1基因在果实成熟过程中呈上调表达模式。An ubiquitin-activating enzyme gene(CpUBA1) was identified in papaya genome by using bioinformatics method. We analyzed the sequence characteristics of this gene,and also studied the expression pattern of this gene during the fruit ripening of papaya. Results showed that the ORF of CpUBA1 was 3 459 bp and encoding 1 152 amino acid residues with an estimated molecular mass of 128.93 ku and a calculated p I of 5.49. Plantm PLoc predicted that CpUBA1 was localized to the nucleus. The results of Real-time q PCR showed that the highest expression level of CpUBA1 gene was tested in leaves,and its lowest expression level occurred in the female flowers. The expression of CpUBA1 was up-regulated during the fruit ripening of papaya.
