摘 要:目的:探讨高迁移率族蛋白1(high mobility group prptein,HMGBI)在促使人肾系膜细胞(human tenal mesangial cell,HRMCs)增殖及分泌纤维化相关因子中的作用。方法:用不同浓度梯度的重组HMGB1刺激体外培养的人肾系膜细胞,以CCK-8测定人肾系膜细胞增殖情况,以ELISA方法检测培养上清液中纤维化相关因子TGF-β1的分泌量,以real-timePCR法测定细胞周期蛋白cyclinD、PNCA蛋白的表达。结果:一定浓度的HMGB1可刺激人肾小球系膜细胞的增殖,细胞内细胞周期蛋白cyclinD、PNCA基因表达增加,细胞培养上清液中TGF-β1浓度增高。结论:HMGBI通过刺激人肾小球系膜细胞增殖并分泌TGF-β1参与了肾脏纤维化过程。Objective To investigate the role of HMGB1 in inducing human renal mesangial ceils {HMCs) to proliferate and secret fibrosis-related cytokines. Methods Recombinant protein HMGB1 (rHMGB1) was incubated with HMCs and then proliferation assay was carried out with CCK-8; cyclinD and PCNA genes were determined with real time PCR; supernate of cultured HRMCs with rHMGB1 was examined for detection of TGF-β1 with ELISA method. Results HMGB1 promoted proliferation of HRMCs; HMGB1 increased expression of cyclinD and PCNA genes involving cell proliferation; rHMGB1 increased secretion of TGF-β1 in supernate of cultured HMCs. Conclusion rHMGB1 was involved in fibrosis of kidney by inducing HRMCs to proliferate and secret fibrosis-related cytokines.
