摘 要:前体蛋白是蛋白质的前体分子,需要在细胞内经各种加工修饰,才能转变成有生物活性的蛋白质。前体蛋白转换酶是在蛋白质分泌通道中对前体蛋白在特定的位点上进行剪切加工的酶。Furin是前体蛋白转换酶大家族中重要的一员,参与前体蛋白加工修饰的过程。Furin的作用底物不仅包括神经肽和肽类激素,还包括许多生长因子、受体、血浆蛋白酶及细菌外毒素等,具有重要的生物学功能。文章对Furin表达加工的过程、结构及Furin相关抑制性药物等方面进行归纳和总结,并且针对Furin加工的前体蛋白和位点进行了汇总。Precursor proteins are precursor molecules of proteins that need to be processed and modified within the cell before they can be turned into biologically active proteins.The precursor protein enzyme is an enzyme that processes the precursor protein at a specific site in the protein secretory channel.Furin is an important member of the large family of proprotein convertases,which is involved in the processing and modification of many kinds of precursor protein.Not only neuropeptides and peptide hormones but also many growth factors,receptors,plasma protease and exotoxin can be the substrate of Furin,which have important biological functions.In this paper,the Furin expression processing,structure and process characteristic and the development status of Furin related inhibitory drugs were summarized.And the precursor proteins and sites of Furin processing were summarized.
