右美托咪定预处理对谷氨酸体外激活的星型胶质细胞和炎症因子分泌的影响

摘　　要：目的:研究右美托咪定(Dexmedetomidine,Dex)预处理对致痛物质谷氨酸体外激活的星型胶质细胞和炎症因子分泌的影响。方法:取新生SD大鼠大脑皮层星型胶质细胞进行原代纯化培养。用25~100 nmol/L Dex预处理原代培养的星型胶质细胞15 min后,再加入有效刺激浓度的谷氨酸共同孵育,24 h后用Western blotting检测星型胶质细胞GFAP蛋白表达水平变化,酶联免疫吸附法检测各组上清液T N F-α、IL-1β含量的变化。结果:纯化培养的星型胶质细胞经10-1000μmol/L谷氨酸刺激24h后,100、1000μmol/L谷氨酸组的星型胶质细胞GFAP的蛋白表达量均比对照组明显增加(P<0.05)。与1000μmol/L谷氨酸组相比,100 nmol/L Dex预处理组星型胶质细胞GFAP蛋白表达和上清液TNF-α、IL-1β含量均明显下降(P<0.05)。结论:一定剂量范围内,Dex预处理可抑制谷氨酸诱导的星型胶质细胞的活性增强和炎症因子释放。Objective：To investigate Effects of dexmedetomidine pretreatment on activation and inflammatory cytokine excretion of astrocytes induced by glutamate in intro.Methods：Astrocytes were taken from cerebral cortex derived from newborn SD rats and incubated.Identify the purity of astrocytes by an astrocytic marker GFAP.Primarily cultured astrocytes were co-cultured with 10~1000 μ mol/L glutamate respectively,and the levels of GFAP were determined by western blot.25~100 nmol/L dexmedetomidine was first added,and 15 min later glutamate with the effective activation concentration was then added.After 24 h of incubation,the levels of GFAP were determined by western blot,and the concentrations of TNF-α,IL-1β by ELISA.Results：Levels of GFAP increased in a concentration-dependent manner after100、1000 μ mol/L glutamate application for 24 h,and 100 and 1000 μ mol/L glutamate significantly increased the expression of GFAP compared with control group（P0.05）.Compared to 1000 μ mol glutamate group,both the expression of GFAP and excretion of TNF-α,IL-1β were significantly attenuated in 100 nmol/L dexmedetomidine pre-administration group（P0.05）.Conclusion：The activation and the inflammatory factor excretion of astrocytes induced by glutamate was significantly attenuated following dexmedetomidine pre-administration.
