灵芝提取物与维生素E对高脂高糖饮食小鼠的协同保护作用研究

摘　　要：目的观察灵芝提取物与维生素E对高脂高糖饮食小鼠的协同保护作用。方法选取6-8周龄的雄性SPF级ICR小鼠60只,随机分为5组,分别为正常组、模型组、灵芝组、维生素E组、灵芝+维生素E复合组;灌胃8周,眼球取血,测定血清中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、总抗氧化能力(T-AOC)等抗氧化指标,以及血液中甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)等血脂相关指标,并观察肝脏组织的病理形态学变化。结果与模型组相比,灵芝提取物可以显著的提高高脂高糖饮食小鼠的HDL-C水平,降低MDA和TC含量;复合组可以显著提高SOD、GSH-Px、T-AOC活力和HDL-C水平,降低MDA、TC、TG和LDL-C含量。肝脏HE染色结果显示,正常组肝细胞排列正常,染色均匀;模型组肝细胞大面积脂肪变性,出现脂肪空泡;实验组肝脏脂肪病变症状较模型组轻,细胞核出现小部分变异。结论灵芝提取物与维生素E复合对高脂高糖饮食小鼠具有一定的协同保护作用。ObjectiveTo observe the synergetic protection of Ganoderma lucidum extract（GLE） and Vitamin E in mice with high fat and high sugar diet. Methods Sixty SPF ICR mice with 6-8 week old were randomly divided into control group, model group, Ganoderma lucidum extract （GLE） group, Vitamin E group and complex group. After pretreatment with gavage for 8 continuous weeks, the mice eyeballs were executed for the sampling of blood in order to detect SOD, MDA, GSH-Px and T-AOC, as well as TG, TC, HDL-C, and LDL-C, and observe the morphologic changes of liver. Results Compared with the model group, the level of HDL-C of rats with high fat high sugar diet was increased and the contents of MDA and TC decreased in Ganoderma lucidum extract （GLE） group; the activities of SOD, GSH-Px, and T-AOC and the level of HDL-C were increased and the contents of MDA, TC, and LDL-C decreased in complex group. Hematoxylin-eosin staining （ HE ） showed the arrangements of the liver cells were trim with even staining. The model group showed a large range of hepatoeyte fatty-change with fat vacuole. The experimental groups HE showed less fat pathological changes in liver compared with the model group only with small area of nuclear variation. Conclusion Ganoderma lucidum extract （GLE） and Vitamin E have synergetic protection on the mice with high fat and high sugar diet.
